摘要
目的探讨溃疡性结肠炎(UC)组织中Toll样受体、前列腺素受体EP2的表达变化及与组织中炎性细胞因子表达的关系。方法选取我院2016年1月至2017年1月收治的100例UC组织(UC组)、50例正常结肠黏膜组织(对照组),采用免疫组化染色检测两组标本中的Toll样受体2(TLR2)、Toll样受体4(TLR4)、前列腺素受体EP2蛋白的表达水平,采用酶联免疫吸附法(ELISA)检测两组标本中的白细胞介素-17(IL-17)、白细胞介素-10(IL-10),并分析TLR2、TLR4、前列腺素受体EP2与IL-17、IL-10的关系。结果 UC组中TLR2、TLR4、前列腺素受体EP2蛋白阳性表达率59. 00%、61. 00%、69. 00%均高于对照组的26. 00%、18. 00%、26. 00%,差异有统计学意义(P<0. 05);活动期、Ⅲ级UC组中TLR2、TLR4、前列腺素受体EP2蛋白阳性表达均显著的高于非活动期、Ⅰ级+Ⅱ级的UC患者,差异有统计学意义(P <0. 05); TLR2、TLR4、前列腺素受体EP2蛋白阳性的UC组织中的IL-17水平高于阴性表达的患者,IL-10表达水平则低于阴性表达的UC组织,差异有统计学意义(P<0. 05)。结论 Toll样受体、前列腺素受体EP2在UC组织表达上调,并且与UC病情进展及炎症反应程度加重有关。
Objective To investigate the expression of Toll-like receptor and prostaglandin receptor EP2 in ulcerative colitis(UC)and their relationship with the expression of inflammatory cytokines in tissues.Methods 100 cases of UC tissue(UC group)and 50 cases of normal colonic mucosa(control group),from January 2016 to January 2017 in our hospital,were concluded.Toll-like receptor 2(TLR2),Toll-like receptor 4(TLR4)and prostaglandin receptor EP2 protein expression levels were detected by immunohistochemical staining,the interleukin-17(IL-17),interleukin-10(IL-10)in two groupwere detected by enzyme-linked immunosorbent assay(ELISA),and the relationship between TLR2,TLR4,prostaglandin receptor EP2 and IL-17,IL-10were analyzed.Results The positive expression rates of TLR2,TLR4 and prostaglandin receptor EP2 protein in UC group were 59.00%,61.00% and 69.00%,and they were higher than 26.00%,18.00% and 26.00% in the control group,statistically significant(P<0.05).The positive expressions of TLR2,TLR4 and prostaglandin receptor EP2 in the active period and grade III UC groups were significantly higher than those in the inactive period and grade I+II UC patients,statistically significant(P<0.05).The level of IL-17 in UC tissues with TLR2,TLR4 and prostaglandin receptor EP2 protein positive expressions was higher than that in patients with negative expression,and the expression level of IL-10 was lower than that of UC tissues with negative expression,statistically significant(P<0.05).Conclusion The expression of Toll-like receptor and prostaglandin receptor EP2 is up-regulated in UC tissues,and is associated with the progression of UC and the severity of inflammatory response.
