基于TCGA数据集分析脑胶质瘤中TEFM基因mRNA的表达及意义 被引量:10

摘要 目的基于癌症基因组图谱(TCGA)数据集分析人脑胶质瘤中线粒体转录延伸因子(TEFM)基因mRNA的表达及其临床意义。方法基因表达谱动态分析(GEPIA)207例非肿瘤脑组织和518例脑胶质瘤组织中TEFM基因mRNA的表达差异。分析518例脑胶质瘤患者的TEFM基因mRNA表达量与临床病理参数和患者预后的相关性,探讨影响脑胶质瘤患者预后的因素。分析脑胶质瘤中TEFM基因与其他线粒体转录调控基因mRNA表达水平的相关性。结果与非肿瘤脑组织相比,人脑胶质瘤组织中TEFM基因mRNA表达水平显著升高(P<0.05)。脑胶质瘤中TEFM基因mRNA表达量与患者年龄、WHO分级、病理类型、头痛史与幕上定位具有显著相关性(P<0.05); TEFM mRNA表达量与脑胶质瘤患者的总体生存率相关性显著(P<0.05)。脑胶质瘤患者年龄、WHO分级、病理类型与幕上定位是影响患者预后的独立因素(P<0.05)。脑胶质瘤中TEFM基因的表达水平与TFAM、TFB1M、TFB2M、MTERF1~4、NRF1基因的表达水平呈显著正相关性(均P<0.01,R=0.59、0.51、0.68、0.70、0.36、0.51、0.51、0.48),与POLRMT基因的表达水平呈显著负相关性(P<0.01,R=-0.18)。结论 TEFM基因mRNA表达量在人脑胶质瘤组织比非肿瘤脑组织中显著升高,且和病理特征具有良好相关性,其表达可能具有预测脑胶质瘤进展和预后的价值。 Objective To investigate expression and significance of human mitochondrial transcription elongation factor (TEFM) in brain glioma on the basis of cancer genome atlas (TCGA) database analysis. Methods Differences of TEFM mRNA expression in 207 samples of nontumorous brain tissues and 518 samples of brain glioma tissues were analyzed by gene expression profiling interactive analysis (GEPIA). Relationships between expression of TEFM mRNA with clinicopathological parameters and prognosis in 518 brain glioma were analyzed. Influential factors of prognosis of patients with brain glioma were analyzed. Relationships between TEFM gene expression with other mitochondrial transcription regulatory genes in brain glioma were analyzed. Results TEFM gene mRNA expression in human brain glioma tissues was significantly higher than that in noncancerous brain tissues ( P <0.05). TEFM mRNA expression was significantly related to age, WHO classification, pathological types, headache history and supratentorial location in brain glioma patients ( P < 0.05). TEFM mRNA expression showed significant correlation with overall survival rate ( P <0.05), but it showed no significant correlation with disease-free survival rate in patients with brain glioma ( P <0.05). Age, WHO classification, pathological types and supratentorial location were independent prognostic factors in brain glioma patients ( P <0.05). TEFM gene expression was positively correlated with expressions of TFAM, TFB1M, TFB2M, MTERF1-4 and NRF1 genes in brain glioma ( P <0.01, R =0.59, 0.51, 0.68, 0.70, 0.36, 0.51, 0.51, 0.48), but it negatively correlated with expression of POLRMT gene ( P <0.01, R =-0.18). Conclusion TEFM gene expression is significantly increased in human brain glioma tissues compared with that in noncancerous brain tissues. Moreover, the expression shows good correlation with clinicopathological characteristics, and the expression may be valuable in prediction of progression and prognosis of brain glioma patients.
