摘要 目的:建立同时测定灵芝三萜类多指标成分的UPLC-MS-MS分析方法。方法:采用液质联用法,对灵芝酸C2、灵芝酸A、灵芝酸G、灵芝酸B、赤芝酸A、灵芝烯酸D进行含量测定。以乙腈(A)-水(B)为流动相,梯度洗脱(0~4 min A的体积分数25%→30%; 4~6 min A的体积分数30%→35%; 6~7. 5 min A的体积分数35%→60%);流速为0. 3 ml·min^(-1),柱温为40℃。采用电喷雾离子源负离子扫描模式进行检测。结果:灵芝中6种成分线性关系、精密度、稳定性、重复性均符合方法学测定要求。加样回收率为96. 5%~100. 2%,RSD为1. 21%~1. 90%(n=6)。结论:利用液质联用法测定各成分含量高效便捷,灵敏度高,可为灵芝的质量评价提供可靠的依据。 Objective:To establish a UPLC-MS-MS method for the simultaneous determination of the multi index components of triterpenoid in Ganoderma lucidum.Methods:The contents of ganoderic acid C2,ganoderic acid A,ganoderic acid G,ganoderic acid B,lucidenic acid A and ganoderma D were determined by using liquid chromatography-mass spectrometry.Acetonitrile(A)-water(B)was used as the mobile phase with gradient elution(0-4 min:A volume fraction from 25%to 30%;4-6 min:A volume fraction from30%to 35%;6-7.5 min:A volume fraction from 35%to 60%).The flow rate was 0.3 ml·min^-1,the column temperature was 40℃.A negative ion scanning mode of electrospray ion source was used.Results:The linear relationship,precision,stability and repeatability of the 6 components of Ganoderma lucidum all conformed to the requirements of methodology.The recoveries were 96.5%-100.2%,RSDs were 1.21%-1.90%(n=6).Conclusion:It is convenient and sensitive to determine the contents of multi index components by using liquid chromatograph.It provides reliable basis for the quality evaluation of Ganoderma lucidum.
