摘要
目的:建立同时测定人血浆中曲唑酮及其代谢产物间氯苯基哌嗪浓度的液相色谱-串联质谱法。方法:采用AB3200Q-TRAP液质联用仪,岛津Inertsil ODS-SP(150 mm×4. 6 mm,5"m)色谱柱,乙腈-7 mmol·L^(-1)乙酸铵水溶液(含0. 1%甲酸)(60∶40)为流动相,乙腈为蛋白沉淀剂处理血浆样品。结果:曲唑酮与内标罗哌卡因的峰面积比在浓度10~5 000 ng·ml^(-1)范围内线性关系良好(r=0. 996 9);间氯苯基哌嗪与内标雷诺嗪在浓度10~1 000 ng·ml^(-1)范围内线性关系良好(r=0. 998 0)。低、中、高浓度质控样品日内、日间精密度RSD和准确度RSD均低于10%。结论:该方法简单快速、准确可靠,可用于曲唑酮及其代谢产物间氯苯基哌嗪血药浓度测定及药动学研究。
Objective:To develop an LC-MS/MS method for the simultaneous determination of trazodone and its active metabolite m-chlorophenylpiprazine in human plasma.Methods:Plasma was analyzed by AB3200 Q-TRAP Mass spectrograph with an Inertsil ODS-SP column(150 mm×4.6 mm,5"m),the mobile phase was acetonitrile and 7 mmol·L^-1 CH3 COONH4(containing 0.1%CH2 O2)(60∶40).The plasma sample was pretreated with acetonitrile.Results:The calibration curve of trazodone showed good linearity over the range of 10-5 000 ng·mL^-1(r=0.996 9).The calibration curve of m-chlorophenylpiprazine showed good linearity over the range of 10-1 000 ng·mL^-1(r=0.998 0).The RSD of intra-and inter-day precisions for low,medium and high concentrations and the RSD of accuracy and the RSD of were less than 10%.Conclusion:The method is simple,accurate and reliable.It can be used for the determination of trazodone and m-chlorophenylpiprazine in human plasma and phamacokinetic studies.
