高效液相色谱法同时测定蒙药森登-4中绿原酸和槲皮素含量 被引量:4

Simultaneous Determination of Chlorogenic Acid and Quercetin in Mongolian Medicine Sendeng-4 by High Performance Liquid Chromatography

摘要 建立反相高效液相色谱法(RP-HPLC)同时测定蒙药森登-4中绿原酸和槲皮素含量的方法.采用超声提取方法作为样品提取方法制备供试品溶液.分析色谱柱为romasil C18(250 mm×4.6 mm,5μm)色谱柱;流动相为甲醇∶水(加0.02%的磷酸)=45∶55;检测波长为360 nm;流速为0.8 mL·min^-1,柱温为室温.结果显示,绿原酸和槲皮素分别在17.6μg·mL^-1~132μg·mL^-1和2.16μg·mL^-1~16.2μg·mL^-1浓度范围内具有良好的线性关系,其相关系数分别为0.9994和0.9996.平均加样回收率测定结果分别为97.3%和98.2%(n=6),相应的RSD值分别为1.2%和1.6%.该方法简便、快速,方法重现性、稳定性和可靠性良好,可作为蒙药森登-4中绿原酸和槲皮素含量测定方法. A high-performance liquid chromatography method(HPLC)for determination of chlorogenic acid and quercetin in Mongolian medicine Sendeng-4 was established. Preparation of sample solution was carried out with ultrasonic extraction. The chromatographic column was romasil C18(250 mm×4.6 mm,5 μm),the mobile phase was methanol:water(0.02% phosphoric acid added)=45∶55,the detection wavelength was 360 nm,the flow rate was0.8 m L·min^-1,and the column temperature was room temperature. There was a good linear relationship between chlorogenic acid and quercetin in the range of 17.6 μg·mL^-1~132 μg·mL^-1 and 2.16 μg·mL^-1~16.2 μg·mL^-1,with correlation coefficients of 0.9994 and 0.9996,respectively. The average recovery was 97.3% and 98.2%,respectively(n=6),and the corresponding RSD was 1.2% and 1.6%,respectively. The method is simple and rapid,with good reproducibility and stability,and can be used for the determination of chlorogenic acid and quercetin in Mongolian medicine sendeng-4.
