摘要
目的:探讨顺铂(DDP)联合长春新碱(VCR)对人结直肠腺癌耐长春新碱细胞(HCT-8/V)增殖、凋亡与自噬的影响。方法:以RT-LCI高内涵技术平台,连续观察120 HCT-8/V细胞的生长规律及连续70 h观察细胞凋亡与自噬。结果:DDP对HCT-8/V有抑制作用,但无剂量依赖关系; VCR对HCT-8/V抑制作用与药物浓度梯度呈正比; DDP、VCR对细胞凋亡的影响没有差异; DDP、VCR单独及联合应用可抑制HCT-8/V自噬。结论:DDP联合VCR应用通过抑制HCT-8/V细胞的增殖与自噬实现耐药性逆转。
Objective:To investigate the effect of cisplatin(DDP) combined with vincristine (VCR)on proliferation,apoptosis and autophagy of human colorectal adenocarcinoma cells resistant to vincristine (HCT-8/V). Methods: RT-LCI high-content technology platform was used to observe the growth of colorectal adenocarcinoma cells for 120 hours and to observe apoptosis and autophagy for 70 hours. Results: DDP has an inhibitory effect on HCT-8/V,but no dose dependence.The inhibitory effect of VCR on HCT-8/V was directly proportional to the drug concentration gradient.There was no difference in the effects of DDP and VCR on apoptosis;DDP,VCR alone and in combination could inhibit HCT-8/V autophagy. Conclusion: DDP combined with VCR application achieves drug resistance reversal by inhibiting proliferation and autophagy of HCT-8/V cells.
