摘要
为了提高土生鳞伞菌丝体的产量,在单因素试验基础上,考察培养温度、摇床转速、培养时间和培养基起始pH对菌丝体干质量的影响,并通过Box-Behnken试验设计和响应面分析法确定土生鳞伞液体发酵最佳工艺条件,对影响土生鳞伞(Native scale umbrella)菌丝生长的关键影响因子最佳水平范围进行研究和探讨。结果表明,土生鳞伞菌丝产量发酵培养条件的最佳组合是:温度25℃,摇床转速180r/min,培养时间9d,pH7,菌丝产量实测值为40.332g/100mL。此研究结果可为土生鳞伞液体发酵的试验生产提供理论依据。
In order to increase the mycelium yield of Umbrella lucidum,the effects of initial pH,culture time,rotating speed of shaker and culture temperature on the mycelium dry mass were investigated on the basis of single factor experiment.The optimum technological conditions of liquid fermentation of Umbrella lucidum lucidum were determined by Box-Behnken design and response surface methodology.The optimum level range of the key factors affecting the mycelial growth of ative scale umbrella was studied and discussed.The results showed that the mycelial yield of native umbrella was fermented.The optimum combination of culture conditions was(100 mL):pH;temperature;rotating speed of shaker;culture time;measured value of m ycelium yield.The mycelium yield of the optimized native umbrella was higher than that before optimization.The results could provide theoretical basis for the experimental production of liquid ferme ntation of native umbrella.
