马齿苋提取液对急性湿疹大鼠皮肤屏障功能的调控作用 被引量:8

摘要 目的探讨马齿苋提取液对皮肤屏障功能的调控作用及其治疗急性湿疹大鼠的可能作用机制。方法 SD大鼠30只,分为正常组、模型组、马齿苋组,各10只。采用二硝基氯苯(DNCB)建立急性湿疹模型,正常组除外。造模成功后,各组分别给予相应药物。用药后,采用皮肤表皮水分丧失测定仪测定各实验组大鼠经皮水分丢失量(TEWL);免疫组织化学法(SP法)染色蛋白酶激活受体2(PAR-2),并采用病理图像分析系统半定量检测其水平;反转录-聚合酶链反应(RT-PCR)测定皮肤人类中间丝聚合蛋白(FLG)基因表达。结果用药后,马齿苋组急性湿疹症状减轻,大鼠皮肤TEWL(3.25±0.47)g·h^(-1)·m^(-2)、PAR-2(0.154 1±0.006 8)μm^2,较模型组显著减少(P<0.05);FLG基因表达值为4.34±1.21,较模型组显著增高(P<0.05)。结论马齿苋治疗急性湿疹的机制之一可能是通过增加FLG基因表达,降低PAR-2水平,减少TEWL,恢复皮肤屏障功能。 Objective To investigate the mechanism of Purslane extracts in regulating the skin barrier function and its possible mechanism in the treatment for rats with atopic eczema.Methods 30 SD rats were randomly divided into the normal group,the model group and the Purslane group with n=10.Acute eczema model was established with dinitrochlorobenzene(DNCB),except the normal group.After successful modeling,the corresponding drugs were given to each group.After medication,the amount of transepidermal water loss(TEWL)in rats was determined by the tester.Immunohistochemical method(SP method)was used to stain protease-activated receptor 2(PAR-2)and detect its level semi-quantitatively by pathological image analysis system.RT-PCR was used to determine gene expression of flilaggrin(FLG)in human skin.Results After medication,the Purslane group presented alleviated acute eczema symptoms.Compared with the model group,the levels of TEWL [(3.25±0.47)g·h^-1·m^-2],PAR-2[(0.154 1±0.006 8)μm^2]in rats skins in the Portulaca group significantly decreased(P<0.01),and the expression of FLG gene(4.34±1.21)increased(P>0.05).Conclusion One of the mechanisms of Purslane for the treatment of acute eczema may be by increasing FLG gene expression,lowering PAR-2 level reducing TEWL,and restoring the skin barrier function.
