摘要
目的研究骨桥蛋白(又称分泌性磷蛋白,spp1)在肺癌中的表达情况及其意义,为今后指导临床治疗、评估患者预后及防治肺癌相关策略及措施提供具有针对性的科学依据。方法将癌症基因图谱(TCGA)数据库中收集的资料进行处理,通过χ~2检验比较3种不同肺癌病理类型突变率的差异;在基因综合表达数据库(GEO)的数据GSE19804中,将癌组织与癌旁正常组织的spp1mRNA进行配对t检验,分析spp1在肺癌中的差异表达情况;在数据GSE30219中,将293例肺癌组织标本先进行单因素log-Rank生存分析,再使用COX模型校正TNM分期因素,进一步通过方差分析或相关分析,分析spp1与TNM分期、性别、年龄之间的关系。结果从TCGA等数据库收集了3 148例肺癌研究数据,得出在鳞状细胞癌中,spp1的突变率的均值为0.01;在肺腺癌中,spp1的突变率的均值为0.00;在小细胞癌中,spp1的突变率的均值为0.02;在Pan-Cancer中的突变率是0.01,不同类型肺癌突变率差异无统计学意义(χ~2=5.64,P>0.05)。从GEO中获得的数据GSE19804,配对t检验结果表明,与癌旁正常组织相比,肺癌组织中的spp1mRNA表达显著上调(t=13.63,df=59,P<0.01);在肿瘤组织中spp1与远处转移(M分期)、淋巴结受累程度和范围(N分期)、年龄呈正相关,与肿瘤大小分级(T分期)和性别无明显关系。结论 spp1在一定程度上影响着肺癌的发生与发展,有望成为肺癌基因诊断、治疗及预后的靶点。
Objective To study the expression and significance of osteopontin(secreted phosphor protein-1,spp1)in lung cancer,and to provide a scientific basis for clinical treatment,prognosis assessment,strategies and measures for prevention and treatment of lung cancer.Methods Data collected in the TCGA database were processed by χ^2 test to compare the difference of mutation rate in three different lung cancer pathological types.In data GSE19804,the expression of spp1 mRNA in cancer tissues and the adjacent normal tissues were analyzed by t-test.In data GSE30219,a single-factor log-rank survival analysis was performed in 293 lung cancer tissue samples,and TNM stage factors were corrected by COX model.The relationships between spp1 and TNM stage,gender and age were further analyzed by ANOVA or related analysis.Results 3 148 cases of lung cancer were collected from TCGA database and the mutation rate of spp1 was 0.01,0.00,0.02,and0.01 in squamous-cell cancer,lung adenocarcinoma,small-cell cancer and Pan-Cancer.The mutation rate was not statistically significant in different types of lung cancer(χ^2=5.64,P>0.05).Data GSE19804 from GEO showed that the expression of spp1 mRNA was up-regulated in cancer tissues compared with that of adjacent normal tissues(t=13.63,df=59,P<0.01).There was a positive correlation between spp1 and distant metastasis(M stage),lymph node involvement and range(N stage),and age,but a negative correlation with tumor size classification(T stage)and gender.Conclusion spp1 played a role in the occurrence and development of lung cancer which is expected to be the target for gene diagnosis,treatment and prognosis of lung cancer.
