摘要
目的建立竹节参Panax japonicus C.A.Mey HPLC指纹图谱,并测定7种核苷类成分的含有量。方法竹节参0.1%氨水提取液的分析采用Venusil MP C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.01%甲酸水,梯度洗脱;体积流量0.8 mL/min;柱温30℃;检测波长254 nm。对结果进行聚类分析和主成分分析。结果 10批样品指纹图谱中有16个共有峰,除S2外相似度均在0.90以上。7种成分在各自范围内线性关系良好(R^2≥0.999 6),平均加样回收率92.74%~98.66%,RSD 2.32%~2.97%。10批样品分为3类。结论该方法简便可靠,重复性好,可用于竹节参药材的质量控制。
AIM To establish the HPLC fingerprints of Panax japonicus C.A.Mey and to determine the contents of seven nucleosides constituents.METHODS The analysis of 0.1%ammonia aqueous extract of P.japonicus was performed on a 30℃thermostastic Venusil MP C18 column(250 mm×4.6 mm,5μm),with the mobile phase comprising of acetonitrile-0.01%formic acid flowing at 0.8 mL/min in a gradient elution manner,and the detection wavelength was set at 254 nm.The results were analyzed by cluster analysis and principal component analysis.RESULTS There were sixteen common peaks in the fingerprints of ten batches of samples,with the similarities of more than 0.90 except S2.Seven constituents showed good linear relationships within their own ranges(R2≥0.999 6),the average recoveries were 92.74%~98.66%with the RSDs of 2.32%-2.97%.Ten batches of samples were divided into three types.CONCLUSION This simple,reliable and reproducible method can be used for the quality control of P.japonicus.
