摘要
目的建立HPLC法测定西红花Crocus sativus L.不同部位中西红花苷-Ⅰ和苷-Ⅱ的含有量。方法西红花不同部位乙醇提取物的分析采用依利特hypersil ODS2柱(4.6 mm×250 mm,5μm);流动相甲醇-水(50∶50);检测波长440 nm;体积流量1.0 mL/min;柱温25℃。结果西红花苷-Ⅰ和苷-Ⅱ分别在1.406~50.48μg/mL(R^2=0.999 9)、1.044~30.65μg/mL(R^2=0.999 9)范围内线性关系良好,平均加样回收率分别为99.32%(RSD=1.15%)、102.16%(RSD=1.59%)。结论该方法准确稳定,重复性好,可用于西红花的质量控制。
AIM To establish an HPLC method for the content determination of croctin-Ⅰand croctin-Ⅱin different parts of Crocus sativus L..METHODS The analysis of ethanol extract of different parts of C.sativus was performed on a 25℃thermostatic Elite hypersil ODS2 column(4.6 mm×250 mm,5μm),with the mobile phase comprising of methanol-water(50∶50)flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 440 nm.RESULTS Croctin-Ⅰand croctin-Ⅱshowed good linear relationships within the ranges of 1.406-50.48μg/mL(R2=0.999 9),1.044-30.65μg/mL(R^2=0.999 9),whose average recoveries were 99.32%,102.16%,with the RSDs of 1.15%,1.59%,respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of C.sativus.
