呼吸道合胞病毒感染单核细胞诱导IDO表达的机制研究 被引量:1

摘要 目的探讨呼吸道合胞病毒(RSV)感染单核细胞诱导IDO表达的分子机制。方法 RSV感染人外周血单核细胞,或预先利用信号通路抑制剂孵育1 h后再进行感染,定量PCR检测IDO的mRNA水平,ELISA检测IDO的蛋白水平。结果与未感染组相比,RSV感染的单核细胞显著上调IDO的mRNA水平和蛋白水平(P <0. 05),并具有时间依赖性;加入TLR3抑制剂或NF-κB抑制剂后可显著抑制RSV感染诱导的IDO水平(P <0. 01);而TLR7/8/9抑制剂、RIG-I抑制剂、PI3K抑制剂、JNK抑制剂和MEK1/2抑制剂对RSV感染诱导的IDO水平则无明显影响。结论 RSV可通过TLR3-NF-κB信号途径诱导单核细胞表达IDO,从而参与其感染相关疾病的发生发展。 Objective To study the mechanism of respiratory syncytial virus( RSV) induced the expression of indoleamine 2,3-dioxygenase( IDO) in monocytes. Methods Monocytes were infected with RSV in vitro,or signal pathway inhibitor were pre-incubated with monocytes for 1 hour and then RSV were added,the level of IDO was detected by RT-PCR and enzyme-linked immunosorbent assay( ELISA).Results After RSV infection,the mRNA and protein levels of IDO by monocytes were significantly increased( P < 0. 05),which was in a time dependent manner. In addition,RSV-induced the expression of IDO by monocytes was significantly decreased after pre-incubated by the TLR3 or NF-κB inhibitor( P < 0. 01),but such effect was not observed when monocytes were pre-incubated with the TLR7/8/9 inhibitor,RIG-I inhibitor,PI3 K inhibitor,JNK inhibitor and MEK1/2 inhibitor. Conclusion RSV can induce the expression of IDO in monocytes via TLR3-NF-κB signal pathway,which may be involved in the development of RSV-related diseases.
