新塔花水提物对动脉粥样硬化模型小鼠IL-1β和TNF-α水平的影响 被引量:2

The Effect of Ziziphora clinopodioides Lam. on the expression of TNF-α and IL-1β in atherosclerosis model mice

摘要 目的研究新塔花水提物对载脂蛋白E基因敲除ApoE(-/-)小鼠血清中炎症因子白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)水平的影响。方法选取40只8 w龄ApoE-/-小鼠和10只同品系C57小鼠,给予高脂饲料喂养94 d建立动脉粥样硬化模型,所有小鼠适应性喂养1 w后,将ApoE-/-小鼠随机分为模型组[0.4 mL/10 g体质量,蒸馏水]、阿托伐他汀组[3 mg/kg体质量,阿托伐他汀]、新塔花高剂量组[1.8 g/kg体质量,水提物]、新塔花低剂量组[0.9 g/kg体质量,水提物],C57小鼠为空白组[0.4 mL/10g体质量,蒸馏水],每组10只,每周记录小鼠体质量变化,并调整用药量。灌胃12 w后所有小鼠禁食不禁水12 h,麻醉后眼球取血,7 500 r/min离心15 min取上液,-80℃冰箱保存备用。称取小鼠体质量并记录;采用HE染色法观察小鼠胸主动脉病理学变化情况;采用酶联免疫双抗夹心法检测小鼠血清中IL-1β和TNF-α含量。结果与模型组比较,阿托伐他汀组、新塔花高剂量组小鼠体质量降低,差异有统计学意义(P<0.05)。与空白组比较,模型组小鼠血清中IL-1β和TNF-α含量水平升高,差异有统计学意义(P<0.05);与模型组比较,新塔花高剂量组、阿托伐他汀组小鼠血清中IL-1β和TNF-α含量水平降低,差异有统计学意义(P<0.05)。结论新塔花具有防治动脉粥样硬化的作用,其机制可能与降低炎症因子IL-1β和TNF-α的表达水平,抑制炎症反应有关,并可能具有一定的药物剂量依赖性。 Objective To observe the effect of water extract of Ziziphora clinopodioides Lam. on serum levels of inflammatory factors interleukin-1 beta(IL-1β) and tumor necrosis factor-alpha(TNF-α) in ApoE(-/-) knockout mice. Methods 40 8-week-old ApoE-/-mice and 10 C57 mice of the same strain were fed with high-fat diet for 94 days to establish atherosclerosis model. After feeding all mice for 1 week, ApoE-/-mice were randomly divided into model group [0.4 mL/10 g BMI, distilled water], statin group 3 mg/kg BMI, statin group [1.8 g/kg BMI,extract], and low-dose group [0.9 g/kg BMI, extract], C57 were blank group [0.4 mL/10 g BMI, distilled water], 10 mice in each group, the weight changes of mice were recorded weekly, and the dosage of C57 was adjusted. After 12 weeks of intragastric administration, all mice were fasting for 12 hours without water. After anesthesia, eyeball blood was collected. After 7 500 r/min for 15 minutes, fluid was taken and stored at-80℃. The thoracic cavity was opened under sterile condition, thoracic aorta was taken and stored in a refrigerator at-80℃ after injection of liquid nitrogen. All mice were weighed and recorded;the pathological changes of thoracic aorta were observed by HE staining;the contents of IL-1β and TNF-α in serum of mice were detected by ELISA. Results Compared with the model group, the body weight of mice in statin group and Ziziphora clinopodioides Lam. high dose group decreased significantly(P<0.05). The results of ELISA showed that the levels of IL-1β and TNF-α in serum of model group were higher than those of blank group(P<0.05). Compared with model group, the levels of IL-1β and TNF-α in serum of Ziziphora clinopodioides Lam. high dose group and statin group were lower(P<0.05). Conclusion Ziziphora clinopodioides Lam. has the effect of preventing and treating atherosclerosis. Its mechanism may be related to reducing the expression of inflammatory factors IL-1β and TNF-α, inhibiting inflammatory response, and may have a dose-dependent effect.
