菊花“红粉”ד延红”杂交种组培快繁技术 被引量:3

摘要 以菊花杂交品种"红粉"×"延红"杂交种子为外植体,研究外植体消毒时间、生长调节剂浓度以及基本培养基浓度对其增殖和生根的影响,旨在建立组培快繁体系。结果表明:0.1%HgCl2消毒8 min,种子发芽率最高为78.3%。在MS+BA 1.0 mg/L+NAA 0.4 mg/L的培养基中,试管苗增殖及生长最佳,株高为9 cm,节数为7.7个。在生根培养中,基本培养基浓度为3/4 MS,当添加IBA 1.0 mg/L时,根生长最佳,根长和根数分别为3.5 cm和15.6条,地上部生长良好,株高和干重均好于其它培养基。将生根的幼苗移栽到栽培基质中(腐殖土∶珍珠岩=3∶1),成活率达95%以上。 To establish a rapid propagation system of ‘Fenghong’בYanhong’ hybrid, seeds of the hybrid as the explant, effects of sterilization time, concentrations of growth regulators and MS medium on shoot proliferation and root growth were determined. The results showed that the maximum seed germination rate reached 78.3% when seeds were sterilized with 0.1% HgCl2 for 8 min. During the proliferation, the optimal culture medium was MS medium supplemented with 1.0 mg/L BA and 0.4 mg/L NAA. Shoot height and number of nodes were 9 cm and 7.7, respectively. During the rooting, root growth was favorable in 3/4 MS medium supplemented with 1 mg/L IBA, where the root length and root numbers per plantlet were 3.5 cm and 15.6, respectively. The growth of the aboveground part, such as shoot height and dry weight was also optimal in 3/4 MS medium+1 mg/L IBA. The survival rate of seedlings was more than 95% when plantlets were transplanted into the cultivation matrix(Humus∶Perlite=3∶1).
