摘要
目的观察血必净注射液通过调控p38 MAPK/NF-κB通路对百草枯中毒大鼠急性肺损伤的保护作用。方法选取SPF级Wistar雄性大鼠,分为5组,除正常组外,其他各组大鼠一次性灌胃百草枯(PQ)溶液染毒。染毒1 h后血必净低、中、高剂量组大鼠分别经尾巴静脉注射含生药0.43、0.86、1.72 g/kg的血必净注射液2 mL,模型组和正常组大鼠注射等剂量0.9%氯化钠注射液。观察大鼠饮食、活动、病理状况,肺组织Bax、Bcl-2和p38-MAPK蛋白及NF-κB及IκB mRNA相对表达量。结果与正常组相比,模型组大鼠肺组织内Bax及Bcl-2蛋白表达上升;与模型组相比,血必净中、高剂量组大鼠肺组织内Bax蛋白表达下降,Bcl-2蛋白表达上升,差异有统计学意义(P <0.05);与正常组相比,模型组大鼠NF-κB m RNA、p38-MAPK蛋白表达上升,IκB mRNA表达下降;和模型组相比,血必净中、高剂量组大鼠NF-κB m RNA、p38-MAPK蛋白表达下降,IκB mRNA表达上升,差异均有统计学意义(P <0.05)。结论血必净注射液通过调控p38 MAPK/NF-κB通路,降低PQ中毒大鼠炎症反应程度,对受损肺组织起到保护作用。
Objective: To analyze the protective effect of Xuebijing Injection on acute lung injury induced by paraquat poisoning in rats by regulating p38 MAPK/NF-κB pathway. Methods: SPF-class Wistar male rats were divided into five groups. Except the normal group,rats in other groups were exposed to paraquat(PQ) solution once. One hour after the exposure,the rats in the low,medium and high doses of Xuebijing Injection groups were injected intravenously with 2 ml of Xuebijing Injection,and the rats in the model group and the normal group were injected with the same amount of normal saline. The diet,activity and pathological conditions of rats were observed. The relative expressions of Bax,Bcl-2 and p38-MAPK proteins,NF-κB and IB mRNA in lung tissues were observed. Results: Compared with the normal group,the expression of Bax and Bcl-2 protein in lung tissue of the model group rats increased. Compared with the model group,the expression of Bax protein in the middle and high dose groups of Xuebijing Injection decreased,and the expression of Bcl-2 protein increased,with statistical significance(P < 0.05);compared with the normal group,the expression of NF-κB mRNA and p38-MAPK protein in the model group increased,and the expression of IκB mRNA decreased. Compared with the model group,the expressions of NF-κB mRNA and p38-MAPK protein in Xuebijing Injection medium and high dose group were decreased,and the expression of IκB mRNA was increased,with statistical significance(P < 0.05). Conclusion: Xuebijing Injection can reduce the degree of inflammatory reaction in rats with PQ poisoning by regulating p38-MAPK/NF-κB pathway,and protect the damaged lung tissue.
