摘要
目的 制备Ce6交联的双孔道介孔二氧化硅纳米粒(MSN/Ce6),对其进行表征和单线态氧产率检测,并考察其对黑色素瘤细胞的杀伤效果。方法 利用透射电镜和紫外-可见分光光度法等手段来表征,采用近红外光电倍增管法检测~1O_2的近红外发光信号,CCK8法评价细胞毒性。结果 制备的双孔道介孔硅纳米粒平均粒径为159.2 nm,电位值为-15.1 m V。UV-vis结果显示MSN成功负载Ce6,近红外结果表明MSN/Ce6的单线态氧产率约为游离Ce6的0.02倍。MSN/Ce6具有显著的细胞毒性,且产生的ROS含量最高,呈浓度依赖性。结论 所制备的共载光敏剂介孔硅纳米粒具有较高的光动力疗效。
Objective The Ce6 cross-linked double-cell mesoporous silica nanoparticles (MSN/Ce6) were prepared, characterized and tested for singlet oxygen yield, and their killing effect on melanoma cells was investigated. Methods It was characterized by transmission electron microscopy and ultraviolet-visible spectrophotometry. The near-infrared luminescence signal of 1O2 was detected by near-infrared photomultiplier tube method, and the cytotoxicity was evaluated by CCK8 method. Results The prepared double-channel mesoporous silicon nanoparticles have an average particle diameter of 159.2 nm and a potential value of -15.1 mV. The UV-vis results showed that MSN successfully loaded Ce6, and the near-infrared results showed that the singlet oxygen yield of MSN/Ce6 was about 0.02 times that of free Ce6. MSN/Ce6 has significant cytotoxicity and produces the highest ROS content in a concentration-dependent manner. Conclusion The prepared co-loading photosensitizer mesoporous silicon nanoparticles have high photodynamic efficacy.
