上调THBS1蛋白的表达对肺腺癌H1975细胞迁移能力的影响 被引量:3

摘要 目的:探讨上调血小板凝血酶蛋白1(THBS1)的表达对肺腺癌H1975细胞体外迁移能力的影响。方法:H1975细胞分为3组,空白对照组不处理,空载体转染组及pc DNA-THBS1转染组分别用脂质体转染空载体pc DNA3. 1和pc DNA3. 1-THBS1真核表达载体,采用划痕实验检测细胞迁移能力,用qRT-PCR、Western blot检测细胞中基质金属蛋白酶(MMP)-2和MMP-9 mRNA、蛋白的表达情况。结果:与空白对照组和空载体转染组相比,pc DNATHBS1转染组H1975细胞迁移能力增强,细胞中MMP-2和MMP-9 mRNA和蛋白的表达升高(P <0. 05)。结论:上调THBS1蛋白可以增强肺腺癌H1975细胞体外迁移能力。 Aim:To study the effects of thrombospondin 1(THBS1)up-regulation on the migration ability of lung ade-nocarcinoma cells in vitro.Methods:H1975 cells were allocated into 3 groups,blank control group was not treated,and empty vector transfection group and pcDNA-THBS1 transfection group were transfected with empty vector and pcDNA3.1-THBS1 eukaryotic expression vector by liposome.The migration ability of cells was detected by cell scratch test.The mR-NA and protein expressions of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)were detected by qRT-PCR and Western blot.Results:Compared with blank control group and empty vector transfection group,the migra-tion ability of cells in pcDNA-THBS1 transfection group was significantly enhanced,and the expressions of MMP-2 and MMP-9 mRNA and protein in the cells were significantly increased(P<0.05).Conclusion:Increasing the expression of THBS1 protein enhances the migration ability of H1975 cells in vitro.
