洛索洛芬钠对急性痛风性关节炎大鼠血清IL-1β和滑膜组织中NF-κB蛋白表达的影响 被引量:5

摘要 目的:探讨洛索洛芬钠对急性痛风性关节炎大鼠血清IL-1β和滑膜组织NF-κB蛋白表达的影响。方法:50只SD大鼠分为正常组(n=12)、模型组(n=12)、治疗组(n=13),阳性对照组(n=13)。模型组、治疗组和阳性对照组大鼠通过采用向膝关节注射尿酸钠来建立急性痛风性关节炎(AGA)模型。治疗组给予0. 3 mg/kg洛索洛芬钠,阳性对照组给予0. 3 mg/kg秋水仙碱,其余两组给予生理盐水灌胃,连续7 d。在造模前后,测量各组大鼠的关节肿胀度;观察其步态变化并进行分级评分;造模24 h后,观察大鼠关节滑膜组织病理形态学变化;用ELISA检测血清中IL-1β的水平,Western blot法检测滑膜组织中NF-κB的蛋白表达。结果:治疗组和阳性对照组各时间点膝关节的肿胀程度较模型组降低(P <0. 05);治疗组和阳性对照组步态水平得分均明显高于正常组,但低于模型组(P <0. 05);治疗组和阳性对照组的NF-κB蛋白表达及IL-1β水平均明显低于模型组(P <0. 05)。结论:洛索洛芬钠可以抑制NF-κB信号通路,减轻机体炎症反应,有效改善实验动物关节肿胀。 Aim:To investigate the effects of loxoprofen sodium on the serum IL-1βlevel and NF-κB expression in synovial tissue in rats with acute gouty arthritis(AGA).Methods:Fifty SD rats were allocated into normal group(n=12),model group(n=12),treatment group(n=13)and positive control group(n=13).AGA model was established by injecting sodium urate into knee joint in model group,treatment group and positive control group.The treatment group was given 0.3 mg/kg loxoprofen sodium,the positive control group was given 0.3 mg/kg colchicine,and the other two groups were given normal saline intragastrically for 7 days.Before and after modeling,the degree of joint swelling was measured,the gait changes were observed and graded.24 hours later,the pathomorphological changes were observed in the joint tissue sections of the rats,the level of IL-1βin serum was detected by ELISA,and the protein expression of NF-κB in synovial tissue was detected by Western blot.Results:The swelling degree of knee joint in the treatment group and positive control group was significantly lower than that in the model group at each time point(P<0.05).The gait scores in the treatment group and positive control group were significantly higher than those in the normal group,but lower than those in the model group(P<0.05).The expression of NF-κB protein and the level of IL-1βin the treatment group and positive control group were significantly lower than those in the model group(P<0.05).Conclusion:Loxoprofen sodium can improve the symptoms of acute gouty arthritis rats.The mechanism may be related to the inhibition of NF-κB signaling pathway and the blockade of inflammatory factor IL-1β.
