摘要
目的:利用生物信息学方法对miR-107的靶基因及功能进行预测,为后续深入研究其生物学功能提供实验验证的理论基础。方法:使用PubMed检索miR-107相关的文章并进行分析总结。通过UCSC基因组浏览器、TargetScan、miRDB、TargetMiner、DAVID等在线工具分析miR-107序列,预测miR-107的靶基因,并对预测的靶基因进行功能富集(GO)分析和信号通路分析。结果:miR-107序列在各个物种间具有高度保守性。文献检索的结果显示,miR-107在一些肿瘤中发挥抑癌基因的作用,而在另一些肿瘤中发挥癌基因的作用。同时miR-107与阿尔茨海默病发病机制有关。GO分析发现 miR-107靶基因涉及的分子功能主要包括泛素结合酶和蛋白丝/苏氨酸激酶活性;生物过程涉及泛素蛋白酶转移和膜内陷;细胞组成包括核浆和核内体(P<0.05)。靶基因涉及的信号通路有mTOR信号通路、胰岛素抵抗和cGMP-PKG信号通路等(P<0.05)。结论:miR-107可能参与多种重要的生物学过程,并作为双向调节因子调控肿瘤的生物学行为。
Objective:To predict the target genes and function of miR-107 using bioinformatics methods and provide a theoretical basis for futher experimental research on its biological function. Methods: The relative studies on miR-107 were retrieved from PubMed. The sequence of miR-107 was got from UCSC Browser. Target genes of miR-107 were predicted by TargetScan, miRDB and TargetMiner, and then analyzed by gene ontology and pathway enrichment analysis. Results: The sequence of miR-107 was highly conserved among various species. The results of literature search showed that, miR-107 plays a role as a tumor suppressor gene in some tumors and as an oncogene in other tumors. Meanwhile, miR-107 is involved in the pathogenesis of alzheimer's disease. GO analysis showed that the molecular functions of the target gene of miR-107 mainly involved in ubiquitin conjugating enzyme binding and protein serine/threonine kinase activity. The biological processes involved membrane invagination. The cell composition included nucleoplasm and endosome(P<0.05). The target genes were involved in mTOR signaling pathway, insulin resistance, and cGMP-PKG signaling pathway(P<0.05). Conclusion: miR-107 may be involved in a variety of important biological processes and acts as a two-way regulator to regulate the biological behavior of tumors.
