理气补肾方对DES诱发勃起障碍大鼠阴茎eNOS蛋白表达的影响

Effects of Liqi Bushen decoction on eNOS protein expression of erectile dysfunction rats induced by DES

目的观察理气补肾方对乙烯雌酚(diethylstilbestrol,DES)诱发的勃起功能障碍Wistar大鼠阴茎功能及一氧化氮合酶(nitric oxide synthase,eNOS)蛋白表达的影响。方法 11周龄雄性Wistar大鼠30只,适应性饲养1周后,分为空白组8只、实验组22只,实验组采用腹腔注射DES诱发大鼠勃起功能障碍,8周后根据大鼠血清睾酮(testosterone,T)及阿扑吗啡(apomorphine,APO)实验筛选勃起功能障碍模型大鼠,并将其按照随机数字表法分为两组,模型组8只,中药组9只。中药组给予理气补肾颗粒剂水溶液灌胃,空白组、模型组给予等体积蒸馏水灌胃。干预4周后,APO实验评价大鼠阴茎勃起功能,腹主动脉取血检测血清促黄体生成素(luteninizing hormone,LH)、卵泡刺激素(follicle stimulating hormone,FSH)及T,HE染色观察大鼠阴茎海绵体组织形态变化,免疫组化观察大鼠阴茎eNOS蛋白表达。结果 与模型组相比,理气补肾中药组显著增加大鼠30分钟内阴茎勃起次数(P<0.05)、缩短勃起潜伏期(P<0.05),升高大鼠血清T水平(P<0.05),增加了阴茎海绵体血窦数量,增加阴茎海绵体eNOS蛋白表达(P<0.05)。结论 理气补肾中药可以显著改善DES诱发勃起功能障碍并增加血清T水平,其机制可能与增加阴茎海绵体eNOS蛋白表达有关。

Objective To observe the effects of Liqi Bushen decoction on penile function and expression of nitric oxide synthase (eNOS) protein of Wistar rats with erectile dysfunction induced by diethylstilbestrol (DES).Methods After 1-week adaptive feeding,30 male Wistar rats of 11 weeks age were divided into blank group (n =8) and experimental group (n =22).The experimental group was given intraperitoneal injection of DES to induce erectile dysfunction.After 8 weeks,according to the serum testosterone (T) and apomorphine (APO) test,the erectile dysfunction model rats were selected and randomly divided into 2 groups,8 in model group and 9 in traditional Chinese medicine (TCM) group.The TCM group was given water solution of Liqi Bushen granules by intragastric administration,while the blank group and the model group were given equal volume distilled water.After 4 weeks of intervention,APO test was used to evaluate the erectile function of the rats,blood samples were taken from the abdominal aorta to detect serum luteinizing hormone (LH),follicular stimulating hormone (FSH) and T,HE staining was used to observe the morphological changes of penile cavernosum of the rats,and immunohistochemistry was used to observe the expression of eNOS protein in the penis.Results Compared with the model group,the number of rat penile erections in 30min was increased significantly in the TCM group (P <0.05),the erection incubation period was shortened (P <0.05),the serum testosterone level in rats was increased (P <0.05),the number of blood sinuses and the expression of eNOS protein in penile cavernosum were increased (P <0.05).Conclusion Liqi Bushen decoction can improve the erectile dysfunction induced by DES and increase the level of serum testosterone,which may be related to the increase of eNOS protein expression in the penile cavernosum.
