摘要
目的探讨CYP2C19在新疆地区维吾尔族、哈萨克族、回族和汉族人群中的基因型及等位基因分布特征,为新疆地区各民族合理用药提供理论依据。方法选取新疆医科大学第一附属医院就诊患者1908例,根据民族分为维吾尔族、哈萨克族、回族和汉族受试组,采用DNA微阵列芯片法检测CYP2C19基因多态性,比较不同民族CYP2C19等位基因、基因型和代谢型的分布差异性。结果新疆地区维吾尔族、哈萨克族、回族和汉族受试组CYP2C19的等位基因、基因型、代谢型分布差异均有统计学意义(P<0.05)。根据等位基因判断:CYP2C19*1占68.11%;CYP2C19*2占27.59%;CYP2C19*3占4.3%;根据基因型判断:快代谢型占47.85%;中间代谢型占40.46%;慢代谢型占11.69%。哈萨克族受试组快代谢型基因频率显著高于其余三个民族;汉族受试组中间型基因频率显著高于其余三个民族;回族慢代谢型基因频率高于其余三个民族,差异均有统计学意义(P<0.05)。结论在新疆地区维吾尔族、哈萨克族、回族和汉族人群中,CYP2C19的等位基因、基因型、代谢型分布有显著差异。维吾尔族和哈萨克族人群以快代谢型为主,回族和汉族人群以中慢代谢型为主,对新疆地区各民族用药有一定指导意义。
Objective To explore the genotype and allele distribution characteristics of CYP2C19 in Uighur,Kazak,Hui and Han populations in Xinjiang and provide theoretical basis for rational use of drugs in ethnic groups in Xinjiang.Methods A total of 1908 patients from the First Affiliated Hospital of Xinjiang Medical University were selected.According to the ethnic group,the Uyghur,Kazakh,Hui and Han subjects were divided into relevant 4 groups.Detection of CYP2C19 gene polymorphism was measured by DNA microarray method;comparision of the distribution differences of CYP2C19 alleles,genotypes and metabolite types in different ethnic group.Results There were significant differences in the allele,genotype and metabolite distribution of CYP2C19 in the Uygur,Kazak,Hui and Han subjects in Xinjiang(P<0.05).According to the allele,CYP2C19*1 accounted for 68.11%;CYP2C19*2 accounted for 27.59%;CYP2C19*3 accounted for 4.3%.According to the genotype,the fast metabolizing type accounted for 47.85%;the intermediate metabolite type accounted for 40.46%;and the slow metabolizing type accounted for 11.69%.The frequency of fast metabolites in the Kazak group was significantly higher than that in the other three ethnic groups;the frequency of the intennediate gene in the Han group was significantly higher than that in the other three ethnic groups;the frequency of the slow metabolite gene in the Hui nationality was higher than that of the other three ethnic groups.All the differences had statistical significance(P<0.05).Conclusion There were significant differences in the allele,genotype and metabolite distribution of CYP2CI9 in the Uyghur,Kazak,Hui and Han populations in Xinjiang.The Uygur and Kazakh populations are mainly fast metabolite,and the Hui and Han populations are mainly intermediate and slow metabolite,which may have certain guiding significance for the use of medicines in various ethnic groups in Xinjiang.
