桑葚提取物主要成分分析及润肠通便作用的观察 被引量:2

Analysis of the main components of mulberry extracts and observation on the function of relaxing bowel

摘要 【目的】研究桑葚提取物和桑葚果汁粉中总黄酮、总花青素含量分布,并比较其润肠通便活性。【方法】分别以芦丁和矢车菊素-3-O-葡萄糖苷(C3G)为标准品,利用紫外分光光度法测定总黄酮和总花青素的含量;以小鼠为研究对象,采用复方地芬诺酯建立小鼠便秘模型,连续给药灌胃7 d,采用酚红测定方法观察受试样品对小肠推进影响。【结果】桑葚提取物中总黄酮含量为79.48%,总花青素含量为27.01%;桑葚果汁粉中总黄酮的含量为0.15%,总花青素的含量为0.13%;给药7 d后,与模型组相比,桑葚提取物高剂量组(1 g/kg)可以显著提高复方地芬诺酯所致便秘模型的小鼠小肠酚红推进率(P<0.05、0.01),而桑葚果汁粉高剂量组(1.5 g/kg)对复方地芬诺酯所致便秘模型的小鼠小肠酚红推进率的提高无显著性。【结论】桑葚提取物中总黄酮和总花青素的含量远远高于桑葚果汁粉,且其润肠通便的效果也明显优于桑葚果汁粉,说明桑葚润肠通便的功效与其黄酮类成分的含量有关。 【Objective】To study the content distribution of total flavonoids and anthocyanins in mulberry extract and mulberry juice powder,and compare their activities of relaxing bowel.【Methods】The contents of total flavonoids and anthocyanins were determined by ultraviolet spectrophotometry using rutin and cypermethrin-3-O-glucoside (C3G) as standard substances respectively,and the samples were gavaged to mice for 7 days.Phenol red test method was used to measure the effect on the intestinal propulsion of constipated mice induced by compound difenoxate.【Results】The content of total flavonoids in mulberry extract was 79.48%.The content of total anthocyanins was 27.01%.The content of total flavonoids in mulberry juice powder was 0.15%,and the content of total anthocyanins was 0.13%.After seven days of administration,compared with the model group,the intestinal phenol red propulsion rate of mice with constipation induced by compound difenoxate could be significantly increased in the high dose mulberry extract group(1 g/kg)(P<0.05,0.01),but the increase in the high dose mulberry juice powder group(1.5 g/kg)was not significant.【Conclusion】The contents of total flavonoids and anthocyanins in mulberry extract are much higher than those in mulberry juice powder,and the effect of mulberry extract on relaxing bowel is far better than that of mulberry juice powder.Therefore,the activity of relaxing bowel in mulberry may be related to the content of flavonoids.
