摘要
目的:探讨前列腺癌血清中长链非编码RNA(long non-codingRNA,lncRNA)HOTAIR的表达和临床意义。方法:收集经病理组织学确诊的前列腺癌患者血清样本62例,正常体检对照血清样本62例,采用TRIzolLS分离血清样本中的RNA;采用qRT-PCR法检测所有血清样本中HOTAIR的表达水平,统计分析血清中HOTAIR水平与前列腺癌临床病理组织参数之间的关系。结果:前列腺癌血清中HOTAIR表达水平高于正常对照组,两组结果之间差异具有统计学意义(P<0.01),TNMⅢ/Ⅳ期的前列腺癌血清中HOTAIR的表达水平高于正常对照组(P<0.05),且Ⅲ/Ⅳ期前列腺癌血清中HOTAIR的表达水平高于Ⅰ/Ⅱ期的表达水平(P<0.05);在有淋巴结转移的前列腺癌患者血清中,HOTAIR的表达水平均高于无淋巴结转移患者(P=0.006);血清中HOTAIR的表达值诊断前列腺癌的灵敏度为69.4%,特异度为71.0%,曲线下面积0.709;且血清中HOTAIR的表达与PSA值呈正相关(P<0.001,r=0.48)。结论:前列腺癌患者血清中HOTAIR的表达水平升高,提示HOTAIR可能参与了前列腺癌的发生发展进程。
Objective:To investigate the expression level and clinical significance of long non-encoded lncRNA HOTAIR in prostate cancer.Methods:Sixty-two serum samples were collected from prostate cancer patients confirmed by histopathology,and another 62 serum samples were obtained from healthy subjects undergone general physical examination.Trizol LS reagent was used to isolate the total RNAs from the serum samples,and quantitative real-time PCR(qRT-PCR) was performed to measure the serum HOTAIR expression.Then the relationship between serum HOTAIR levels and clinical pathological parameters were statistically analyzed.Results:Serum HOTAIR levels were significantly increased in patients of prostate cancer compared with the control subjects (P<0.01).Prostate cancer patients in TNM stage Ⅲ/Ⅳ had significantly higher serum HOTAIR levels than the healthy subjects (P<0.05),and the patients in TNM stage Ⅲ/Ⅳ had higher serum levels of HOTAIR than those in TNM stage Ⅰ/Ⅱ(P<0.05).Patients with lymph node metastasis had significantly higher serum of HOTAIR levels than those without lymph node metastasis ( P= 0.006).The sensitivity and specificity by serum HOTAIR in the diagnosis of prostate cancer were 69.4% and 71.0%,respectively.The area under the receiver-operating characteristic curve (AUC) was 0.709.Serum HOAIR expression was positively correlated with PSA(P<0.001,r= 0.48).Conclusion:The serum HOTAIR level can be significantly increased in patients with prostate cancer,suggesting that HOTAIR expression may be involved in the pathogenesis of prostate cancer.
