摘要
目的:探究人晶状体上皮细胞(LEC)内色素上皮衍生因子(PEDF)的表达与糖尿病视网膜病变的关系。方法:收集白内障患者术中所取晶状体前囊膜,将其分为非糖尿病(NDM)组、无糖尿病视网膜病变(NDR)组、非增殖型糖尿病视网膜病变(NPDR)组和增殖型糖尿病视网膜病变(PDR)组。免疫组织荧光法及蛋白质免疫印迹技术分别用于检测PEDF蛋白在LEC中的分布和表达。结果:PEDF蛋白主要分布于LEC胞浆中。随着糖尿病视网膜病变的出现和病情进展,LEC中PEDF蛋白的表达水平显著增加(P<0.01)。结论:PEDF的表达水平与糖尿病视网膜病变的发生和发展呈正相关,表明人晶状体本身有助于调节虹膜新生血管。
Objective: To investigate the expression of pigment epithelium-derived factor (PEDF) in human lens epithelial cells (LEC) and explore their relationships with diabetes to evaluate the mechanism of lens regulating neovascularization. Methods: Lens anterior capsules attached with LEC were collected from cataract surgeries in patients with or without diabetes, and grouped as following: non-diabetes mellitus (NDM) group, no diabetic retinopathy (NDR) group, non-proliferative diabetic retinopathy (NPDR) group and proliferative diabetic retinopathy (PDR) group. The expression of PEDF in all groups was determined by Western blot and immunofluorescence assay. Results: PEDF protein was detected in LEC and mainly distributed in the cytoplasm. The results showed that, the level of PEDF protein expression in LEC was significantly increased with the appearance and aggravation of diabetic retinopathy ( P <0.01). Conclusion: The level of PEDF protein expression is positively correlated with the appearance and aggravation of diabetic retinopathy. This suggests that the human lens itself may help regulate iris neovascularization.
