摘要
目的卵泡抑素样蛋白l(follistatin-like protein 1,FSTL1)是一种分泌性糖蛋白,在人类多种肿瘤中出现异常表达。本研究旨在探讨FSTL1在宫颈癌中的表达情况及与宫颈癌高危型人乳头状瘤病毒(high-risk human papilloma virus, HR-HPV)感染的关系。方法收集2015-04-13-2017-05-24广西壮族自治区人民医院妇科66例宫颈癌组织样本和 21 例正常宫颈组织样本。采用实时荧光定量 PCR (fluorogenic quantitative reverse transcriptase polymerase chainreaction, RT-qPCR)检测样本中 FSTL1 mRNA 的表达量,巢式多重 PCR (nested multiplex polymerase chain reaction,NMPCR)检测66例宫颈癌组织样本中HPV-16、18、31、45、52和58分型,分析FSTL1 mRNA的表达量与HPV感染的关系;将宫颈癌HeLa细胞株分为实验组、阴性对照组和空白对照组,实验组转染HPV-18E6/E7小干扰RNA(smallRNA interference, siRNA),阴性对照组转染阴性对照siRNA,空白对照组不做处理,每组设置3个复孔,RT-qPCR技术检测HPV-18E6/E7 siRNA转染前后宫颈癌HeLa细胞株中FSTL1 mRNA的表达情况。结果 FSTL1 mRNA在66例宫颈癌组织样本中相对表达量的中位数(极差)为0. 059(0.004,0. 789),在21例正常宫颈组织相对表达量的中位数(极差)为1. 051(0. 517,1. 539),差异有统计学意义,Z= 6. 765,PV0. 001;FSTLl mRNA在HPV感染阳性组中相对表达量的中位数(极差)为0. 057(0. 004,0. 698),在HPV感染阴性组中相对表达量的中位数(极差)为0. 226(0. 030,0. 789),差异有统计学意义,Z= 3. 071,P<0. 001;FSTL1 mRNA高表达组(27. 27%)的HPV感染率低于低表达组(93. 94%),差异有统计学意义,才=5. 345, P = 0. 021;HeLa 细胞转染 HPV-18E6 siRNA 后,HPV-18E6 mRNA(0. 269 + 0. 037,F=79. 946, PV0. 001)的相对表达量下调,FSTL1 mRNA的相对表达量为2. 922 士 0. 239,较阴性对照组(0. 978 + 0. 076)及空白对照组(1. 000 + 0. 029)上调,差异有统计学意义,F= 175. 347, P<0. 001;HeLa细胞转染HPV-18E7 siRNA 后,HPV- 18E7 mRNA(0. 335± 0. 040, F= 50. 442, PV0. 001)表达量下调,FSTL1 mRNA 的相对表达量为3.798 + 0. 059,较阴性对照组(0. 906土0. 058)及空白对照组(1. 001 士0. 060)上调,差异有统计学意义,F=2 330. 835,P<0. 001。结论 FSTL1 mRNA在宫颈癌组织中低表达,FSTL1 mRNA的表达量与宫颈癌HPV感染有关。
OBJECTIVE Follistatin-like protein 1 ( FSTL1) is a secretory glycoprotein, which is abnormally expressed in a variety of human tumors. The recent research on FSTL1 in cervical cancer has rarely been reported. This study aimed to investigate the expression FSTL1 in cervical cancer and and its relationship with high-risk human papillomavirus. METHODS The expression of FSTL1 mRNA of 66 cases of cervical cancer tissues and 21 cases of normal cervical cancer tissue samples collected from the Department of Gynecology Guangxi Zhuang Autonomous Region People’s Hospital from 2015-04-13 - 2017-05-24, was detected and analyzed by fluorogenic quantitative reverse transcriptase polymerase chain reaction(RT-qPCR). The nested multiplex polymerase chain reaction(NMPCR) was used to detect and analyze the classification of HPV-16,18,31,45,52,58 in the above 66 cervical cancer tissues samples. Analysis of the relationship between the expression of FSTL1 mRNA and HPV infection. The cervical cancer HeLa cell line was divided into experimental group,negative control group and blank control group. The experimental group was transfected with HPV-18E6/E7 small RNA interference(siRNA),the negative control group was transfected with negative control siRNA,the blank control group was not treated,and each group was set with 3 duplicate wells. RT-qPCR was used to detect the expression of FSTL1 mRNA in cervical cancer HeLa cell line before and after HPV-18E6/E7 siRNA transfection. RESULTS The median( range) relative expression of FSTL1 mRNA in 66 cervical cancer tissues was 0. 059(0. 004,0. 789),and the median (range) relative expression in 21 normal cervical tissues was 1. 051(0. 517,1. 539),the difference was statistically significant( Z=-6. 765,PV0. 001). The median(range) relative expression of FSTL1 mRNA in the HPV-positive group was 0. 057(0. 004,0. 698),and the median(range) relative expression in the HPV-infected group was 0. 226(0.030,0.789), the difference was statistically significant(Z=- 3. 071, P<C0. 001). The HPV infection rate in the high expression group of FSTL1 mRNA (27. 27%) was lower than that in the low expression group (93. 94%),and the difference was statistically significant(x^2=5. 345 , P = 0. 021). After HeLa cells were transfected with HPV-18E6 siRNA, the expression level of HPV-18E6 mRNA(0. 269土0. 037 ,F= 79. 946 ,PV0. 001) was down-regulated. The relative expression of FSTL1 mRNA was (2. 922±0. 239) higher than that of the negative control group (0. 978 zt 0. 076) and the blank control group (1. 000±0. 029). The difference was statistically significant(F- 175. 347,PV0. 001). After HeLa cells were transfected with HPV-18E7 siRNA,the expression level of HPV-18E7 mRNA (0. 335dz0. 040, F= 50. 442, PV0. 001) was downregulated. The relative expression of FSTL1 mRNA was 3. 798 + 0. 059, which was higher than that of the negative control group (0. 906 ±0. 058) and the blank control group (1. 001 ±0. 060). The difference was statistically significant (F = 2 330. 835,PV0. 001). CONCLUSION The expression of FSTLlmRNA is down-regulated in cervical cancer tissues and it is related to HPV infection in cervical cancer.
