摘要
目的:观察助阳补肺除痹颗粒对肺纤维化大鼠肺组织SOD活性、MDA含量的影响。方法:SPF级Wistar大鼠,按体重随机分为6组,每组30只动物。分别为假手术组、模型组、醋酸泼尼松组及助阳补肺除痹颗粒高、中、低组,采用博来霉素复制肺纤维化大鼠模型,假手术组大鼠以等量生理盐水代替盐酸博来霉素。各组大鼠自建模后第2天起每天灌胃给予相应药物1次,分别于造模后第7天、第14天和第28天给药后1h,每组随机选取10只动物,处死后剖取右侧肺组织,采用分光光度法检测肺组织SOD活力、MDA含量。结果:肺组织中SOD活性比较,假手术组高于模型组(P<0.05),醋酸泼尼松组动物及助阳补肺除痹颗粒高剂量组均高于同期模型组(P<0.05),助阳补肺除痹颗粒中剂量组、低剂量组在第28天时高于同期模型组(P<0.05)。肺组织中MDA含量比较,假手术组低于模型组(P<0.01),醋酸泼尼松组动物及助阳补肺除痹颗粒高剂量组低于同期模型组(P<0.05),助阳补肺除痹颗粒中剂量组低于同期模型组(P<0.05),低剂量组在第28天时低于同期模型组(P<0.05)。结论:助阳补肺除痹颗粒能够通过增加肺组织中SOD活性,降低MDA的释放,抑制肺纤维化的进程。
Objective:To observe the effect of Zhuyang Bufei Chubi granule on SOD activity and MDA content in lung tissue of rats with pulmonary fibrosis.Methods:SPF Wistar rats were randomly divided into 6 groups according to body weight,30 animals in each group were sham-operated group, model group.prednisone acetate group.The high,middle and low dose of Zhuyang Bufei Chubi granule group.The model of pulmonary fibrosis was induced by bleomycin.In the sham operated group,the equivalent saline was used to replace bleomycin hydrochloride.The rats in each group were given the corresponding drugs once a day from 2 day after modeling,after modeling 7th day,14th day and the 28 th day respectively,10 animals were randomly selected in each group.The right lung tissue was dissected and the SOD activity and MDA content in lung tissue were detected by spectrophotometry.Results:Compared with the model group,the SOD activity in the lung tissue of the sham operated group was higher than that of the model group (P< 0.05).Prednisone acetate group and high dose group of Zhuyang Bufei Chubi granule were higher than the model group (P<0.05).Middle dose group and low dose group on the 28 day was higher than that of the model group (P<0.05).Compared with the model group,the MDA content in the sham operated group was lower than that in the model group (P<0.01).Prednisone acetate group and high dose group were lower than model group (P<0.05).Middle dose group was lower than the model group (P<0.05),and the low dose group was lower than the model group on the 28 th day (P<0.05).Conclusion:Zhuyang Bufei Chubi granule can increase SOD activity in lung tissue,decrease the release of MDA content and inhibit the process of pulmonary fibrosis.
