摘要
目的:探讨替吉奥治疗大鼠胃癌前病变的效果以及对Notch信号通路蛋白Notch1和Jagged1表达情况的影响。方法:将60只大鼠随机取等分3组:对照组、模型组、替吉奥治疗组。用100μg/ml的MNNG溶液灌胃,制作大鼠胃癌模型。模型建立成功后治疗组给予替吉奥治疗药物,对照组以及模型组给予等体积的生理盐水。PCR法检测胃癌前病期组织Notch信号通路Notch1和Jagged1的基因表达水平;Westernblot检测Notch1和Jagged的蛋白表达量。结果:PCR检测结果显示,Notch1和Jagged1在对照组中仅有少量的表达,与对照组相比,模型组的表达出现了显著增高(P<0.05),相比与模型组,替吉奥治疗组表达有明显下降,仍高于对照组的表达(P<0.05);Westernblot检测结果表明,Notch1和Jagged1在对照组中蛋白仅有微量的基础表达,与对照组相比,模型组的两蛋白表达水平明显地上升(P<0.05),同模型组相比,替吉奥治疗组的两蛋白表达均有所下降,但仍高于对照组(P<0.05)。结论:替吉奥治疗可显著降低大鼠胃癌前病变组织Notch1和Jagged1蛋白的表达。
Objective: To explore the effect of S-1 (Tegafur,Gimeracil and Oteracil Porassium Capsules)treatment on the expression of Notch signaling pathway protein Notch1 and Jagged1 in rat gastric precancerous lesions. Methods: 60 healthy male SD rats were randomly and equally divided into three groups:control group,model group,and S-1 treatment group.The rat gastric cancer model was established by the intragastric administration of 100 μg/ml MNNG solution.Rats with gastric precancerous lesions were treated with S-1 or saline.The mRNA levels of Notch1 and Jagged1 were detected by PCR.The protein levels of Notch1 and Jagged1 were detected by Western blot. Results: PCR showed that Notch1 and Jagged1 mRNA levels were significantly higher in the gastric tissues of model group than that of control group;S-1 treatment could partially decrease the mRNA levels of these two genes( P <0.05).Consistent with the PCR result,Western blot result showed that S-1 treatment decreased the levels of Notch1 and Jagged1 protein in rat gastric precancerous lesions. Conclusion: S-1 treatment can decrease Notch1 and Jagged1 expression in rat gastric precancerous lesions.
