CDX2和SOX4在胃癌组织中的表达及临床意义 被引量:4

摘要 目的:探讨尾型同源盒转录因子2(caudal type homeobox transcription factor 2, CDX2)和Y 染色体上的性别决定区相关高迁移率族盒蛋白4(sex-dertermining region of Y chromosome related high mobility group box 4, SOX4)在胃癌组织中的表达及其临床意义。方法:采用Western Blot法检测CDX2 蛋白和SOX4 蛋白在50 例胃癌及其远癌胃组织中的表达,分析两者与胃癌临床病理特征之间的关系,Pearson 相关系数分析CDX2 蛋白、SOX4 蛋白在胃癌组织中表达的相关性;免疫荧光法检测蛋白细胞内定位。结果:免疫荧光通过镜下观察,CDX2 阳性信号在胃癌组织中主要位于细胞核,SOX4 阳性信号也见于细胞核。Western Blot 结果提示,CDX2 蛋白、SOX4 蛋白在胃癌组织中高表达,在远癌胃组织中低表达(P<0.05)。CDX2 蛋白表达与胃癌浸润深度、胃癌的TNM 分期、有无淋巴转移有关(均P<0. 05),与肿瘤分化程度、肿块大小、年龄和性别等无关(P>0.05)。SOX4 蛋白表达与胃癌有无淋巴转移和TMN分期有关(P<0. 05),与肿瘤分化程度、浸润深度、肿块大小、年龄和性别等无关(P>0.05)。Pearson 相关性分析提示在胃癌组织中CDX2 和SOX4 蛋白表达呈负相关(r=-0.476, P<0.05)。结论:CDX2 低表达和SOX4 高表达与胃癌病情进展有关,CDX2 和SOX4 在胃癌组织中表达呈负相关。联合检测CDX2和SOX4 的表达可作为反映胃癌临床病理学特点新的分子病理标志物。 Objective : To explore the relationship between caudal type homeobox transcription factor 2(CDX2), sex-dertermining region of Y chromosome related high mobility group box 4(SOX4) biological features in gastric cancer( GC) patients. Methods : The expression of CDX2 and SOX4 protein was detected by Western Blot in 50 surgically dissected GC and matched normal gastric mucosa tissue samples. The relationship between CDX2 and SOX4 protein expression with clinicopathological characteristics of gastric cancer was investigated. The correlation of CDX2 and SOX4 protein expression in GC tissues was analyzed by Pearson correlation coefficient. Moreover, the positions of CDX2 and SOX4 protein in GC were detected using immunofluorescence technique. Results : Immunofluorescence showed that CDX2 positive signal was mainly located in the nucleus of GC, and SOX4 positive signal was also found in the nucleus. Western Blot suggested that CDX2 and SOX4 protein were highly expressed in GC tissues and low expressed in normal noncancerous tissue(all P<0.05). CDX2 protein was significantly correlated with the depth of invasion, lymph node metastasis and TNM stages(all P<0.05), but not correlated with the degree of differentiation, tumor size, age and gender(all P>0.05). The expression of SOX4 protein was significantly correlated with lymphatic metastasis and TNM stage(all P<0.05), but not correlated with differentiation, depth of invasion, size, age and gender(all P>0.05). Pearson correlation analysis showed that CDX2 expression is negatively correlated with SOX4 expression in gastric cancer( r=-0.476, P<0.05). Conclusions : CDX2 and SOX4 proteins may be related to invasion and metastasis of gastric cancer. E xpression o f C DX2 w as n egatively c orrelated w ith S OX4 e xpression i n g astric c ancer. T he d etection o f C DX2 and SOX4 protein may be helpful in evaluating the clinicopathological parameters and prognosis in gastric carcinoma a patient.
