β-TrCP1在非霍奇金淋巴瘤细胞黏附耐药中的作用

摘要 目的:探讨β转导重复包含蛋白(β-transducin repeat containing protein 1,β-TrCP1)对非霍奇金淋巴瘤(non-Hodgkin's lymphoma, NHL)细胞黏附耐药性的影响。方法:采用Western Blot法检测NHL细胞黏附到骨髓基质细胞后β-TrCP1的表达水平,钙黄绿素检测法检测β-TrCP1 对NHL 细胞黏附率的影响,CCK8、TUNEL 检测干扰β-TrCP1 后对NHL 细胞耐药性的影响。结果:β-TrCP1 蛋白在NHL 细胞黏附到骨髓基质细胞后表达升高,且干扰β-TrCP1 的表达能明显下调NHL 细胞的黏附率。将NHL 细胞黏附到骨髓基质细胞并加入米托蒽醌后,干扰β-TrCP1 的表达能明显抑制NHL 细胞活性并增加凋亡细胞数量。结论:干扰β-TrCP1 能抑制NHL 细胞黏附介导的耐药。 Objective : To investigate the role of β-transducin repeat containing protein 1(β-TrCP1) on drug resistance of non-Hodgkin's lymphoma(NHL) cells. Methods: The expression of β-TrCP1 was detected in NHL cells adhered to HS5 by Western Blot. The adherence rate and the effect of drug resistance was detected by Calcein and CCK8 or TUNEL, respectively after the redution of β-TrCP1expression in NHL cells. Results :β-TrCP1 was overexpressed in NHL cells adhered to HS5. Silence of β-TrCP1 significantly down-regulated adherence rate of NHL cells. In addition, interference of β-TrCP1 obviously inhibited the viability of NHL cells and increased the apoptosis cells when NHL cells were adhered to HS5 treated with mitoxantrone. Conclusion : Interfering β-TrCP1 inhibited the progress of cell adhesion mediated drug resistance in NHL.
