摘要
在前期研究中发现赤子爱胜蚓(Eisenia fetida)的甲醇提取物具有体外抗菌活性。将干燥蚯蚓用甲醇浸泡、超声波提取、有机溶剂萃取粗提物,以抗菌活性分析为导向,采用正相和反相色谱相结合的方法,富集E.fetida中的非极性活性分子群Fr.2-5。运用TLC、HPLC-DAD、~1H NMR和GC-MS对Fr.2-5的物质组成进行了相互佐证分析。结果表明:Fr.2-5对Vibrio Parahemolyticus ATCC17802和Chromobacterium violaceum ATCC12472有一定的抑制活性,活性部位主要由(不)饱和脂肪酸(酯)和少量甾体类化合物组成。
Earthworms are excellent animal feedstuff,important environmental guardians and traditional Chinese medicine.In our previous work,we have discovered that methanol extract of Eisenia fetida showed antibacterial activity in vitro.In this paper,the active components were analyzed.The earthworms were soaked with methanol,extracted by ultrasonic method,partitioned with organic solvent to obtain the extract.Under the guidance of antibacterial activity-oriented method,positive and reverse phase chromatography were used to get the fraction of Fr.2-5.Fr.2-5 was the non-polar mixture with antibacterial activity against Vibrio Parahemolyticus ATCC17802 and Chromobacterium violaceum ATCC12472.TLC,HPLC-DAD,1H NMR and GC-MS were used to elucidate its compositioa It was a mixture of(unsaturated)fatty acids(esters)and a small number of steroids.This study provides reference for the resource utilization of earthworms.
