摘要
为研究白茅内生菌Chaetomium globosum WQ固体发酵产物的化学成分,以1H-NMR谱和TLC为指导,采用硅胶柱层析、Sephadex LH-20凝胶柱层析、反相柱层析和高效液相色谱等方法对C.globosum WQ固体发酵产物乙酸乙酯相的化学成分进行分离纯化,并采用MTT法测定化合物对氧糖剥夺(OGD)模型大鼠星形胶质细胞的体外神经保护活性。结果表明:共分离获得5个细胞松弛素类化合物cytoglobosin Ab(1)、chaetogloKosin W(2)、chaetoglobosin V(3)、chaetoglobosin Vb(4)和cytoglobosin I(5);体外活性测定发现,cytoglobosin Ab(1)能明显降低OGD引起的大鼠星形胶质细胞死亡,且呈浓度依赖性。这一研究提示cytoglobosin Ab(1)可能是治疗新生儿缺氧缺血性脑病(NHIE)潜在的神经保护剂。
To investigate the chemical constituents from the solid culture of endophyte Chaetomium globosum WQ associated with Imperata cylindrical,guided by 1H-NMR spectrum and TLC,isolation and purification of the ethyl acetate extract of the solid culture of C.globosum WQ was carried out through silica gel column chromatography,gel filtration over Sephadex LH-20,ODS column chromatography and HPLC.Then,in vitro neuroprotective effect of compounds a-gainst oxygen glucose deprivation(OGD)-induced damage in rat astrocytes was evaluated by MTT method.Five cytochalasan compounds cytoglobosin Ab(1),chaetoglobosin W(2),chaetoglobosin V(3),chaetoglobosin Vb(4)and cytoglo-bosin I(5)were obtained.In vitro neuroprotective effect assay showed that cytoglobosin Ab(1)could attenuate OGD induced cell death in rat astrocytes in a dose-dependent manner obviously,which indicated that cytoglobosin Ab(1)would be a potential neuroprotective agent in treatment of Newborn Hypoxic Ischemic Encephalopathy(NHIE).
