以壳聚糖金纳米粒子为载体构建Ch- GNPs/β6复合物的相关研究 被引量:2

摘要 通过凝聚过程形成Ch-GNPs,以其为载体,搭载整合素β6质粒,形成Ch-GNPs/β6复合物。利用紫外可见分光光度计,透射电子显微镜(TEM)检测复合物的性状。进一步通过电泳、WB和X-gal实验检测复合物的结合力及对MC3T3-E1细胞的转染能力。结果表明Ch-GNPs/β6复合物性状稳定并能有效在MC3T3-E1细胞中进行转染。 The Ch-GNPs were conjugated with the pcDNA-β6 through a coacervation process. As the carrier, with integrin αvβ 6 plasmid, Ch-GNPs/β 6 compound was formed. The size and surface characterization of Ch-GNPs were examined using UV-vis spectroscopy and TEM. The DNA conjugation and transfection capacity of Ch-GNPs were studied by agarose gel electrophoresis, β-galactosidase staining and immunoblotting.The study suggests that the chitosan and the acid chloride alloy through the simple condensation process can form Ch-GNPs compounds.The Ch-GNPs with the right granularity can be transfected into MC3 T3-E1 cells.
