四磨汤口服液中3种成分人血浆蛋白结合率测定 被引量:3

Determination of Human Plasma Protein Binding Rates of Three Main Components in Simotang Oral Liquid

摘要 目的研究四磨汤口服液中去甲异波尔定、柚皮苷、新橙皮苷与人血浆蛋白的结合率。方法采用平衡透析法测定四磨汤口服液中去甲异波尔定、柚皮苷、新橙皮苷3种成分与人血浆蛋白的结合率,用高效液相色谱法(HPLC)测定透析内、外液中3种成分的质量浓度,计算血浆蛋白结合率。结果去甲异波尔定、柚皮苷和新橙皮苷的线性关系、精密度、提取回收率和稳定性等均符合方法学要求;在低、中、高3个浓度的四磨汤口服液中,去甲异波尔定与人血浆蛋白结合率分别为(73.09±1.79)%、(70.95±0.67)%、(79.07±3.43)%;柚皮苷与人血浆蛋白结合率分别为(72.44±1.45)%、(74.73±0.16)%、(80.83±2.69)%;新橙皮苷与人血浆蛋白结合率分别为(75.71±1.49)%、(76.03±0.39)%、(79.84±3.17)%。结论四磨汤口服液中去甲异波尔定、柚皮苷、新橙皮苷属于中高度结合型成分。 Objective To study the protein-binding rates of Norisoboldine,Naringin,Neohesperidin in Simotang oral liquid with human plasma proteins.Methods Equilibrium dialysis method was conducted to determine the binding rates of 3 components in Simotang oral liquid in vitro.An HPLC method for the determination of the 3 comp on ents both in inner and outer dialysis was established and then the protein-binding rates of the comp on ents with human plasma proteins were calculated.Results Linear relationship,precision,extraction recovery and stability of Norisobokline,Naringin,Neohesperidin all complied with the methodological requirements.At the low,middle and high concentrations of Simotang oral liquid,the human plasma protein-Hindi ng rates were(73.09±1.79)%,(70.95±0.67)%and(79.07±3.43)%for Norisoboldine,(72.44±1.45)%,(74.73±0.16)%and(80.83±2.69)%for Naringin,(75.71±1.49)%,(76.03±0.39)%and(79.84±3.17)%for Neohesperidin.Conclusion Norisoboldine,Naringin,Neohesperidin in Simotang oral liquid has middle and high capacities in binding to human plasma proteins in vitro.
