摘要
该研究以菌体OD600 nm值为响应值,采用响应面法对木醋杆菌NUST4.2的种子扩大培养条件进行优化。通过Plackett-Burman试验设计筛选主要影响因素,在此基础上,采用最陡爬坡路径法逼近最大响应值区域,最后利用响应面分析法确定最优培养条件。结果表明,主要影响因素为搅拌转速、通气量、消泡剂用量;木醋杆菌NUST4.2的种子扩大培养的最优条件为装液量70%,初始pH值为6.0,搅拌转速257 r/min,消泡剂用量0.054%,罐压0.2 MPa,通气量1.72 Nm3/h。在此最优条件下培养36 h,OD600 nm值达到0.70。
Taking the OD600 nm of cell as the response value, the cultivation conditions of the Acetobacter xylinum NUST4.2 seeds were optimized by response surface methodology. The main influencing factors were selected by Plackett-Burman experimental design. On this basis, the maximum response value region was approached by the steepest ascent experiments, and the optimal conditions were determined by response surface methodology. The results showed that the main influencing factors were stirring speed, ventilation and defoaming agent addition. The optimal enlarging cultivation conditions of A. xylinum NUST4.2 seeds were liquid loading volume 70%, initial pH value 6.0, stirring speed 257 r/min, defoaming agent addition 0.054%, fermentor pressure 0.2 MPa, ventilation 1.72 Nm^3/h. The OD600 nm value was 0.70 after cultured for 36 h under the optimal conditions.
