摘要
低温大气压等离子体(CAP)近年来作为一种新兴的抗癌手段逐渐为人们所熟知。三阴性乳腺癌在多种乳腺癌亚型中恶性程度最高,由于缺少有效的治疗手段导致预后很差。发现等离子体激活的培养基(PAM)能够选择性抑制三阴性乳腺癌细胞增殖和迁移能力,而对正常细胞和非三阴性乳腺癌细胞影响较小,同时PAM能够抑制MDAMB231中原本高度激活的JNK的磷酸化程度。因此推测累积的基因组突变和高度激活的MAPK/JNK信号通路使三阴性细胞更容易被PAM杀死。研究提出了CAP杀伤作用与乳腺癌亚型之间的相关性,并且为探索CAP选择性杀死三阴性细胞的机制提供了前期基础。
Cold atmospheric plasma(CAP) has become a promising onco-therapeutic approach in recent years. Triple negative breast cancer is most aggressive among various breast cancer subtypes and has worst prognosis due to lack of effective therapy. We found that plasma-activated medium(PAM) selectively inhibited the proliferation and migration of triple negative breast cancer cells while causing little effect on other subtypes, and PAM selectively inhibited the phosphorylation of JNK in MDAMB231. We suggested that it was the more genome mutation and hyper-activated MAPK/JNK pathway in triple negative breast cancer cells that made them more sensitive to CAP. Our work revealed the relation between CAP and breast cancer subtypes and provides a preliminary basis for exploring the mechanism of CAP selectively killing triple negative breast cancer cells.
