聚酰胺树脂纯化金鸡毛草叶总黄酮及其体外抗氧化作用研究 被引量:7

摘要 用单因素试验优化聚酰胺树脂纯化金鸡毛草叶总黄酮工艺,采用单因素试验以维生素C为对照品,考察总黄酮纯化液的抗氧化活性。结果表明:树脂最佳纯化工艺为上样pH 3、洗脱液乙醇的浓度70%、上样体积1.2 BV、上样流速1.5 BV/h,在此条件下纯化后总黄酮的保留率为54.44%,纯化后样液纯度提高至4.52倍;抗氧化活性方面,在干浸膏浓度20~100μg/mL范围内对1,1-二苯基-2-三硝基苯肼自由基(DPPH)清除率为20.79%~50.66%,在100μg/mL浓度下对羟自由基(OH)的清除率为3.58%,在3.6~18.2μg/mL浓度范围内对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS+)清除率为28.71%~82.14%,表明金鸡毛草叶总黄酮具有一定的抗氧化活性。为金鸡毛草的深入开发、产业附加值的提高提供参考依据。 The process of total flavonoids from extract of Rooster maocao leaves purified by polyamide resin was optimzed by single factor test.Vitamin C was used as reference substance to investigate the antioxidant activity of total flavonoids.The results showed that the optimum purification process was pH of 3,ethanol concentration of 70%,volume of 1.2 BV and flow rate of 1.5 BV/h.Under these conditions,the retention rate of total flavonoids was 54.44%,and the purity of the puri fied sample was increased to 4.52 times.The scavenging rate of DPPH was 20.79%-50.66%in the range of 20-100μg/mL for antioxidant activity,the OH clearance rate was 3.58%at 100μg/mL,and the ABTS+clearance rate was 28.71%-82.14%in the range of 3.6-18.2μg/mL,indicating that the total flavonoids of Rooster maocao had certain antioxidant activity.A reference for further development and industrial added value of Rooster maocao was provided.
