摘要 在考察碳源、氮源、碳氮比、pH、发酵时间、发酵温度及金属离子对甲磺酸去铁胺产率影响的单因素实验基础上,引入星点设计—响应面优化法,考查碳氮比、发酵时间及发酵温度3个因素对甲磺酸去铁胺产率的影响,并建立各因素与产率关系的数学回归模型.得出结果:甲磺酸去铁胺最佳发酵工艺为是以蔗糖为碳源,天冬酰胺为氮源,碳氮质量比为8∶1,30℃,48h,培养基初始pH为7. Based on a single factor experiment including carbon source,nitrogen source,carbon to nitrogen ratio,pH,fermentation time,fermentation temperature and metal ion on bacteria growth and deferoxamine mesylate yield,the star point design response was introduced in order to optimize the fermentation process of deferoxamine mesylate produced by bacteria.The surface optimization method was used to examine the effects of carbon and nitrogen ratio,fermentation time and fermentation temperature on the bacteria growth and the yield of deferoxamine mesylate.The three dimensional effect diagram was drawn by design expert 8.0 software to investigate the optimum process conditions.The optimum fermentation conditions of deferoxamine mesylate were consistent with the optimal fermentation conditions of the bacteria growth,in detail:sucrose as the carbon source and asparagine as the nitrogen source,carbon nitrogen mass ratio was 8∶1,30℃,48 h,the initial pH of the medium was 7.
