摘要
目的:采用HPLC法建立野生和栽培白及的指纹图谱,同时建立白及中天麻素、militarine的含量测定方法,比较野生和栽培白及中天麻素和militarine的含量。方法:色谱柱为ZORBAX SB-Aq(250 mm×4.6 mm,5μm);以流动相乙腈(A)-0.1%磷酸水溶液(B)梯度洗脱,检测波长为220 nm,柱温30℃,流速1.0 mL/min,进样体积10μL。结果:野生白及指纹图谱共检测出18个共有峰,栽培白及指纹图谱共检测出16个共有峰,二者峰形整体一致。野生白及中天麻素与militarine平均含量较栽培白及高。结论:该研究建立的白及HPLC指纹图谱和2种指标成分定量测定分析方法简便、准确、重现性好,可用于白及药材的质量控制。
Objective:To establish fingerprints of wild and cultivated Bletilla striata tuber by HPLC and a method for determining the contents of gastrodin and militarine,and to compare the contents of gastrodin and militarine of wild and cultivated Bletilla striata.Methods:The HPLC condition was listed as follows:Agilent ZORBAX SB-Aq column(250 mm×4.6 mm,5 μm)was used;the mobile phase was acetonitrile(A)-0.1% phosphoric acid(B)with gradient elution,detection wavelength was 220 nm,column temperature was 30 ℃,flow rate was 1.0 mL/min,injection volume was 10 μL.Results:A total of 18 common peaks were detected in wild Bletilla striata fingerprint.A total of 16 common peaks were detected in cultivated Bletilla striata fingerprint.The peak shapes of the two fingerprints were the same.The average contents of wild Bletilla striata of gastrodin and militarine were higher than that of cultivatied Bletilla striata.Conclusion:HPLC fingerprints and the quantitative determination methods of the two indicators are simple,accurate,and reproducible,and can be used to control the quality of medicinal materials of Bletilla striata.
