摘要
目的:探讨熟地黄对幼年注意缺陷多动障碍模型大鼠行为学的干预作用。方法:将30只SHR随机分为模型组、专注达(2 mg/kg)阳性对照组、熟地黄(2.4 g/kg)组,另设WKY大鼠为正常对照组,每组10只。正常对照组和模型组均以0.5%羧甲基纤维素钠(CMC-Na)溶液灌胃,各组均按2 mL/kg在大鼠夜周期分2次灌胃,连续给药4 w。采用开场实验、高架十字迷宫、水迷宫评估大鼠自发性行为、冲动行为及学习记忆功能。结果:开场实验中,给药4 w时,与模型组比较,专注达组与熟地黄组总运动距离、直立次数、中心区域进入次数均显著降低(P<0.05),且熟地黄组直立次数、理毛次数显著少于专注达组(P<0.05);高架十字迷宫示,与模型组比较,专注达组和熟地黄组总运动距离、进入开臂次数比、开臂停留时间、中心区域停留时间均显著降低(P<0.05),且熟地黄组中心区域停留时间显著短于专注达组(P<0.05);水迷宫结果示,与模型组比较,专注达组与熟地黄组在第2~5天隐蔽站台潜伏期显著缩短,Morris水迷宫潜伏期均显著缩短(P<0.05),穿越平台次数、目标象限停留时间均显著增加(P<0.05)。结论:熟地黄能减少幼年注意缺陷多动障碍模型大鼠自发性行为、冲动行为,提高其学习记忆能力。
Objective:To observe the impact of Rehmannia glutinosa on behavior of ADHD model rats.Methods:30 spontaneous hypertension rats(SHR)were randomly divided into model group,Methylphenidate Hydrochloride(MPH)(2 mg/kg)positive control group and Rehmannia glutinosa group(2.4 g/kg/day)with ten rats in each group.Ten wistar-kyoto(WKY)rats were selected as normal control group.0.5% CMC-Na solution was administered to the normal group and model group(2 mL/kg).All rats were treated for 4 w.The open field test,elevated plus maze and morris water maze were performed to evaluate the effects on spontaneous,impulsive behavior and learning-memory ability.Results:In the opening experiment,compared with the model group,the total distance of movement,vertical times and central region entry times of MPH group and Rehmannia glutinosa group were significantly reduced when the drug was administered at 4 w(P<0.05);the numbers of erect times and hairdressing times of Rehmannia glutinosa group were significantly lower than that in MPH group(P<0.05).Overhead cross maze experiment showed that compared with model group,the total movement distance,enter the open arms ratio,open arms dwell time and center area residence time of MPH group and Rehmannia glutinosa group were significantly lower(P<0.05),and the duration of stay in the central region of Rehmannia glutinosa group was significantly short than that in MPH group(P<0.05).Water maze experiment results showed that compared with model group,the incubation periods of MPH group and Rehmannia glutinosa group were decreased significantly(P<0.05),the platform number and the target quadrant residence time were significantly increased(P<0.05).Conclusion:Rehmannia glutinosa can reduce spontaneous behavior and impulsive behavior of juvenile attention deficit hyperactivity model rats,and improve their learning and memory ability.
