摘要
目的:对胡桃楸及同属植物胡桃进行本草考证,并对其进行DNA条形码鉴定。方法:通过查阅《食疗本草》《开宝本草》《本草纲目》等本草古籍及满药传统文献,并结合第四次全国中药资源普查进行实地调查,对胡桃楸及其同属植物胡桃进行名称、入药部位及功效的考证;将46份样品进行ITS序列的扩增与测序,且从GenBank中下载相关植物的ITS序列。所有序列经DNAstar软件拼接,采用MEGA 6.0软件分析处理,基于K2P模型构建Neighbor-Joining(NJ)系统发育树。结果:胡桃楸是典型的满药材资源;胡桃楸同属植物胡桃为张骞出使西域引入种;对16份胡桃楸样品测序得ITS序列长度均为648 bp,通过NJ树构建可将胡桃楸、胡桃、枫杨等48个样品有效鉴定。结论:该研究为胡桃楸的应用历史、资源开发及鉴定提供了依据。
Objective:To perform the herbal textual research of Juglans mandshurica and Juglans regia in the same genus,and to identify the DNA barcode.Methods:The names medicinal parts and effects of Juglans mandshurica and Juglans regia were identified by conducting field surveys combined with referring to Herbal Materia Medica,Kaibao Materia Medica,and Compendium of Materia Medica and other traditional herbal medicine books and traditional Chinese medicine resources.46 samples were subjected to amplification and sequencing of the ITS sequence,and the ITS sequences of related plants were downloaded from GenBank.All sequences were spliced by DNA star software and analyzed by using MEGA 6.0 software.The Neighbor-Joining(NJ)phylogenetic tree was constructed based on the K2 P model.Results:Juglans mandshurica was a typical Manchu medicinal material resource;Juglans regia was introducted from the western region.The ITS sequences of 16 samples of Juglans mandshurica were all 648 bp in length,through the construction of NJ tree,48 samples of Juglans mandshurica,Juglans regia,Pterocarya stenoptera etc were effectively identified.Conclusion:The study provides the basis for the application history,resource development and identification of Juglans mandshurica.
