摘要
目的:建立醋三棱主要化学成分的分析方法。方法:采用超高效液相色谱-飞行时间质谱联用(UPLC-TOF-MS)法对醋三棱70%乙醇提取物化学成分进行快速分析鉴定;色谱柱:Waters ACQUITY^(TM) BEH C_(18)(100 mm×2.1 mm,1.7μm);以0.05%甲酸水溶液(A)-0.05%甲酸乙腈溶液(B)梯度洗脱;流速0.4 mL/min;柱温30℃;进样量2μL。质谱条件:AB Sciex Triple TOF 5600质谱仪;ESI源,运用正负离子模式分析。结果:在优化的液质条件下,通过分析正负离子质谱信息及元素组成,并结合相关文献,共鉴定出醋三棱中33个化合物。结论:通过UPLC-TOF-MS技术,建立了一种简单、快速、高效的醋三棱化学成分的分析鉴定方法。
Objective:To analyze the chemical composition of vinegar Sparganii Rhizoma by UPLC-TOF MS.Methods:UPLC-TOF MS was used to analyze and identify the 70% ethanol extract in vinegar Sparganii Rhizoma.The mobile phase was composed of acetonitrile with 0.05% formic acid-water with 0.05% formic acid in gradient mode,the HPLC column was Waters ACQUITYTM BEH C18(100 mm×2.1 mm,1.7 μm),the separation were performed by gradient elution with the flow rate of 0.4 mL/min,the column temperature was 30 ℃,and the injection volume was 2 μL.Mass spectrometry conditions:AB Sciex Triple TOF 5600 mass spectrometer with ESI ion source was used to detect the fragments,the ion mode scan were positive and negative.Results:Under optimized LC/MS condition,a total of 33 chemical compounds of vinegar Sanleng were identified by analyzing the information of positive and negative ion mass spectrometry.Conclusion:A simple,rapid and efficient analytical method with the usage of UPLC-TOF-MS is established to identify the multi-compounds of vinegar Sparganii Rhizoma.
