芪蛭三七汤对心肌梗死后大鼠缺血心肌血管新生及VEGF mRNA表达的影响

Effects of qizhi sanqi decoction on angiogenesis and VEGF mRNA expression in the rats with myocardial ischemia after myocardial infarction

目的观察芪蛭三七汤对心肌梗死后大鼠缺血心肌血管新生及缺血心肌血管内皮生长因子(Vascular endothelial growth factor,VEGF)mRNA表达的影响。方法结扎大鼠的左冠状动脉,造成急性心肌梗死模型,随机分为模型组(阴性对照组)、麝香保心丸组(阳性对照组)4 mg/kg、芪蛭三七汤组16 ml/kg,同时设假手术组。8周后检测各组大鼠缺血心肌中灰度值、微血管密度(Microvessel density,MVD)及血管内皮生长因子mRNA的表达。结果芪蛭三七汤组及麝香保心丸组的VEGF mRNA表达值均要明显高于模型组,差异有统计学意义(P<0.05),芪蛭三七汤组略低于麝香保心丸组,差异无统计学意义(P>0.05);芪蛭三七汤组与麝香保心丸组的微血管密度(MVD)比模型组明显增多(P<0.05)。结论芪蛭三七汤可促进心肌梗死后大鼠缺血心肌血管新生,作用机理可能与促进VEGF mRNA表达有关。

Objective To observe the effects of qizhi sanqi decoction on angiogenesis and VEGF mRNA expression in the rats with myocardial ischemia after myocardial infarction.Methods The left coronary artery was ligated in the rats to prepare the model of acute myocardial infarction.They were randomized into a model group(negative control group),a shexiang baoxin pills group(positive control group,4 mg/kg)and a qizhi sanqi decoction group(16 ml/kg).Additionally,a sham-operation group was prepared.8 weeks later,the myocardial grey value,microvessel density(MVD)and VEGF mRNA expression were determined in each group.Results In the qizhi sanqi decoction group,the positive control group and the model group,MVD in the marginal zone of myocardial infarction was increased obviously as compared with the sham-operation group(P<0.05).MCD in the qizhi sanqi decoction group was higher obviously than the model group(P<0.05)and lower than the positive control group,but without significant differences(P>0.05).Conclusion Qizhi sanqi decoction promotes angiogenesis in the rats with myocardial ischemia after myocardial infarction and its underlying mechanism is related to the promotion of VEGF mRNA expression.
