补中益气汤不同配伍对脾虚证大鼠尿液代谢组学的影响

Effects of Different Combinations of Buzhong Yiqi Decoction on Urine Metabolomics in Rats with Spleen Deficiency Syndrome

目的:运用代谢组学方法研究补中益气汤及其配比组方对脾气虚证大鼠尿液代谢表型变化的影响,探讨补中益气汤"益气升阳"配伍的科学内涵。方法:采用核磁共振氢谱(~1H-NMR)分析大鼠尿液的代谢谱,应用主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)筛选与脾虚证及补中益气汤作用相关的代谢标志物,比较补中益气汤及其配比组方治疗后大鼠与脾虚证大鼠尿液代谢谱的差异。结果:模型组的代谢谱发生了明显的变化,从中筛选出与脾虚证相关的代谢产物8种,补中益气汤对其中6种代谢物水平变化有较大的逆转作用,作用趋势优于其配比组方。结论:补中益气汤原方对脾虚证大鼠的代谢恢复调节作用优于去"升麻-柴胡"和倍"升麻-柴胡"的配比组方,原方中"益气升阳"的药物剂量配比更为合理。

Objective:To study the effects of Buzhong Yiqi decoction and its proportion on the change of urine metabolic phenotype in the rats with spleen qi deficiency syndrome by using metabolomics method,and to explore the scientific connotation of Buzhong Yiqi decoction′s "Benefiting Qi and Raising Yang" combination.Methods:Nuclear magnetic resonance spectroscopy(1H-NMR)was used to analyze the metabolic spectrum of rat urine.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to screen the related metabolic markers of spleen deficiency syndrome and Buzhong Yiqi decoction.The differences in urine metabolism profiles between rats treated with Buzhong Yiqi decoction and its altered prescriptions were compared.Results:The metabolic profiles of model group changed significantly,and eight metabolites related to spleen deficiency syndrome were screened out.Buzhong Yiqi decoction had a greater reversal effect on the changes of six metabolites levels,and the action mentioned above was better than that of its altered prescriptions.Conclusion:The regulatory effect of Buzhong Yiqi decoction to recover the metabolites of rats with spleen deficiency syndrome is better than that of its altered prescriptions in Buzhong Yiqi decoction without cimicifuga and bupleurum and Buzhong Yiqi decoction with doubled cimicifuga and bupleurum.It′s more reasonable of the dose ratio in the original Buzhong Yiqi decoction,which accounts for "Benefiting Qi and Raising Yang" compatibility.
