摘要
目的:主要研究重度子痫前期中人胎盘绒毛膜板动脉(human placental chorionic arteries,HPCA)经典瞬时受体电位通道6(transient receptor potential canonical 6,TRPC6)的表达情况,探讨TRPC6的表达与该疾病的相关性。方法:采用实时荧光定量PCR法(qPCR)及蛋白免疫印记法(Western Blotting)来检测TRPC6在正常孕妇组、早发型重度子痫前期组及晚发型重度子痫前期组的mRNA及蛋白表达情况。结果:早发型重度子痫前期TRPC6的mRNA的相对表达量为2.820 0 ±1.028 7,晚发型重度子痫前期TRPC6的mRNA的相对表达量为1.918 3 ± 0.896 4,差异有统计学意义(P < 0.05)。在正常孕妇组TRPC6的蛋白表达量为0.510 4 ± 0.091 6,TRPC6在早发型重度子痫前期的蛋白表达量是1.373 0 ± 0.171 2,在晚发型重度子痫前期的表达量为0.911 4 ± 0.128 0,差异有统计学意义(P < 0.05)。结论:TRPC6在重度子痫前期孕妇组中的mRNA及蛋白表达水平高于正常孕妇组,故猜测TRPC6可能参与了重度子痫前期的发病过程,早发型重度子痫前期孕妇组的mRNA及蛋白表达水平高于晚发型重度子痫前期孕妇组,且重度子痫前期的发病时间越早,该通道的蛋白表达量越高。
Objective: To investigate the expression of transient receptor potential canonical 6 (TRPC6) in human placental chorionic arteries in severe preeclampsia and the correlation between the expression and the disease. Methods: Real-time quantitative PCR and Western blotting were used to measure the mRNA and protein expression of TRPC6 in the normal pregnant women group, the early-onset severe preeclampsia group, and the late-onset severe preeclampsia group. Results: The relative mRNA expression of TRPC6 was significantly different between the early-onset group and the late-onset group (2.820 0 ± 1.028 7 vs 1.918 3 ± 0.896 4, P < 0.05). The protein expression of TRPC6 was significantly different between the normal group, the early-onset group, and the late-onset group (0.510 4 ± 0.091 6 vs 1.373 0 ± 0.171 2 vs 0.911 4 ± 0.128 0, P < 0.05). Conclusion: The mRNA and protein expression of TRPC6 in the severe preeclampsia groups was higher than that in the normal pregnant women group. Therefore, it is speculated that TRPC6 may be involved in the development of severe preeclampsia. The mRNA and protein expression of TRPC6 in the early-onset severe preeclampsia group was higher than that in the late-onset severe preeclampsia group, and the earlier the onset, the higher the protein expression.
