摘要
目的 探讨血浆循环miRNA-21是否与心房颤动(房颤)的发生有关,并初步探讨循环miRNA-21参与房颤发生的机制。方法 选择2014年3月至9月于心血管内科住院的房颤患者112例作为房颤组,非房颤患者99例作为对照组,用qRT-PCR检测其血浆循环miRNA-21的表达;并采用mirFocus和starBase等生物信息学软件对miRNA-21进行生物信息预测。结果 房颤患者与对照组间年龄、性别、基本病史和生化检查等差异无统计学意义(P>0.05);房颤患者血浆循环miRNA-21表达水平较对照组明显升高(P<0.01);ROC曲线分析发现血浆循环miRNA-21预测房颤的曲线下面积为0.70(P<0.01);生物信息学预测发现miRNA-21的KEGG信号通路主要有MAPK信号通路和TGF-beta信号通路,其靶基因主要是TGFB2、TGFBR1、TGFBR2。结论 血浆循环miRNA-21可能参与房颤的发生,并且可以作为房颤的生物标志物;而其有可能通过调控MAPK信号通路和TGF-beta信号通路而导致房颤的发生。
Objective To investigate whether plasma circulating miRNA-21 is related to the occurrence of atrial fibrillation,and to explore the mechanism of circulating miRNA-21 in the occurrence of atrial fibrillation. Methods 112 patients with atrial fibrillation who hospitalized in cardiovascular department from March to September 2014 were selected as atrial fibrillation group,and 99 patients without atrial fibrillation as control group.The expression of circulating microRNA-21 in plasma was detected by qRT-PCR,and bioinformatics software such as mirFocus and starBase were used to perform the biological information prediction of miRNA-21. Results There was no statistically significant difference in age,gender,basic history and biochemical examination between atrial fibrillation patients and the control group( P >0.05);the expression level of circulating miRNA-21 in patients with atrial fibrillation was significantly higher than that in the control group( P <0.01);ROC curve analysis found that area under curve of plasma circulating miRNA-21 which was used to predict atrial fibrillation was 0.70( P <0.01);bioinformatics prediction discovered that the KEGG signaling pathway of miRNA-21 mainly consisted of MAPK signal pathway and TGF-beta signal pathway,and its target genes were mainly TGFB2,TGFBR1 and TGFBR2. Conclusion Plasma circulating miRNA-21 may participate in the occurrence of atrial fibrillation,and can be used as biomarker of atrial fibrillation.And it may lead to the occurrence of atrial fibrillation by regulating MAPK signaling pathway and TGF-beta signaling pathway.
