阿尔茨海默病患者淀粉样斑块的对比增强磁共振成像与淀粉样变的血清标志物的相关性研究 被引量:1

摘要 目的探究阿尔茨海默病患者淀粉样斑块的对比增强磁共振成像与淀粉样变的血清标志物的相关性。方法,选取108例AD患者(研究组)和105位健康老年人(对照组),获取血清后提取到总RNA,并进行Hiseq测序。筛选出AD患者血清miRNAs拷贝数大于10且变化2倍以上。应用McDonald法对qRT-PCR结果进行标准化,内参为miR-cel-39,相对表达量采用2-ΔΔCt进行表征。采用Pearson相关系数评估各变量之间的相关性,得到阿尔茨海默病患者淀粉样斑块的对比增强磁共振成像与淀粉样变的血清标志物的相关性和相关系数。结果,对照组脑组织中未见淀粉样斑块,而AD患者组的颅脑总淀粉样斑块体积为1363.76mm3。首先在小批量样本即初筛组中得到14种miRNA,然后运用qRT-PCR对初筛出的14种miRNA进行复筛。六个miRNA在AD患者血清中的表达量低于对照组血清(P<0.05)。颅脑总淀粉样斑块体积与血清中miR-98-5p、miR-885-5p、miR-483-3p、miR-342-3p、miR-191-5p和let-7d-5p表达量呈正相关(P<0.05)。结论,阿尔茨海默病患者淀粉样斑块的对比增强磁共振成像与淀粉样变的血清标志物miR-98-5p、miR-885-5p、miR-483-3p、miR-342-3p、miR-191-5p和let-7d-5p表达量呈正相关。 Objective To explore the correlation between contrast enhanced magnetic resonance imaging (MRI) and amyloidosis serum markers in amyloid plaques of patients with Alzheimer ’s disease(AD). Methods,A total of 108 AD patients(study group) and 105 healthy controls(control group) were selected. After obtaining the serum, the total RNA was extracted and sequenced by Hiseq. The miRNAs copy number of AD patients was more than 10 and the change was more than 2 times. All qRT-PCR results were standardized according to McDonald method, and miR-cel-39 was used as internal reference. The relative expression was characterized by 2- Delta Ct. The correlation between variables was evaluated with Pearson correlation coefficient, and the correlation and correlation coefficient of the contrast enhanced MRI with the serum markers of amyloidosis in AD patients were obtained. Results,No amyloid plaques were found in control group, while the total amyloid plaques in the AD group were 1 363.76 mm 3. First of all, 14 kinds of miRNA were obtained in the small batch sample, namely the initial screening group, and then qRT-PCR was used to screen the initial 14 miRNAs.The expression of the six miRNAs in serum of AD patients was significantly lower than that of control group ( P<0.05). The volume of total amyloid plaques in the brain was positively correlated with the expression levels of miR-98-5p, miR-885-5p, miR-483-3p, miR-342-3p, miR-191-5p and let-7d-5p in serum( P<0.05). ConclusionThe contrast enhancement of amyloid plaque in patients with AD has a significant positive correlation with the serum markers of amyloidosis, miR-98-5p, miR-885-5p, miR-483-3p, miR-342-3p, miR-191-5p and let-7d-5p.
