抗氧化剂对烧伤家兔应激期炎性反应的保护作用及抗氧化损伤功能的影响 被引量:2

摘要 目的观察硫辛酸对烧伤家兔应激期炎性反应的保护作用及抗氧化损伤功能的影响。方法新西兰大白兔20只,造成深Ⅱ度烧伤动物模型,随机分为给药组和对照组各10只。给药组采用硫辛酸40 mg/kg灌胃,对照组不给药,比较2组烧伤前、烧伤后血浆内毒素水平、细菌培养阳性率及血清TNF-α、IL-6、IL-8、丙二醛(MDA)、一氧化氮(NO)、超氧化物歧化酶(SOD)水平。结果烧伤前及烧伤后1 d 2组血浆内毒素水平比较差异无统计学意义(P>0.05);烧伤后3 d、5 d给药组血浆内毒素水平均低于对照组(P<0.05)。2组烧伤前、烧伤后1 d、7 d细菌培养阳性率比较差异均无统计学意义(P>0.05);给药组家兔烧伤后3 d、5 d细菌培养阳性率低于对照组(P<0.05)。烧伤前2组血清TNF-α、IL-6及IL-8水平比较差异均无统计学意义(P>0.05);烧伤后2组血清TNF-α、IL-6及IL-8水平均升高,但给药组烧伤后3 d、5 d、7 d TNF-α、IL-6及IL-8水平均低于对照组,差异均有统计学意义(P<0.05)。烧伤前2组MDA、NO和SOD水平比较差异无统计学意义(P>0.05)。血清MDA水平在烧伤后3 d达峰值,烧伤后5 d降低;血清NO水平在烧伤后3 d达最低值,5 d后有所升高;血清SOD水平在烧伤后3 d、5 d均降低。但给药组烧伤后3 d、5 d血清MDA水平低于对照组,血清NO、SOD水平高于对照组,差异均有统计学意义(P<0.05)。结论烧伤家兔给予硫辛酸治疗能明显控制炎性反应,减少脂质过氧化作用,减轻机体氧化损伤。 Objective To observe the protective effect of lipoic acid on the inflammatory response of burned rabbits during stress period and the effect of anti-oxidative damage.Methods 20 New Zealand white rabbits,causing deep Ⅱdegree burn animal models,and they were randomly divided into drug-administered group(10 rats) and the control group(10 rats).The drug-administered group was administered with lipoic acid 40 mg/kg,the control group was not administered,the levels of plasma endotoxin,bacterial culture positive rate and the levels of serum TNF-α,IL-6,IL-8,malondialdehyde(MDA),nitric oxide(NO) and superoxide dismutase(SOD) were compared between the two groups before and after burn.Results There was no significant difference in plasma endotoxin levels between the two groups before and after burn(P>0.05);The plasma endotoxin levels in the drug-administered group were lower than those in the control group at 3 and 5 days after burn(P<0.05).There was no significant difference in the positive rate of bacterial detection between the two groups before burn and 1 d and 7 d after burn(P>0.05);The positive rate of bacterial detection at 3 d and 5 d after burn in the drug-administered group was lower than that of the control group(P<0.05).There were no significant differences in serum levels of TNF-α,IL-6 and IL-8 between the two groups before burn(P>0.05).The levels of serum TNF-α,IL-6 and IL-8 in the two groups after burn were observed(P>0.05),but the levels of TNF-α,IL-6 and IL-8 in the drug-administered group were significantly lower than in the control group at 3,5 and 7 days after burn(P<0.05).There was no significant difference in the levels of MDA,NO and SOD between the two groups before burn(P>0.05).Serum MDA levels peaked at 3 days after burn and decreased at 5 days after burn;Serum NO levels reached the lowest value at 3 days after burn and increased after 5 days;Serum SOD levels decreased at 3 days and 5 days after burn.However,the levels of serum MDA in the drug-administered group were lower than those in the control group at 3 and 5 days after burn,and the levels of serum NO and SOD were higher than those in the control group(P<0.05).Conclusion Burning rabbits treated with lipoic acid can significantly control the inflammatory response,reduce lipid peroxidation and reduce oxidative damage.
