Advances in research of Tau protein leading to cognitive dysfunction

摘要 认知功能障碍是临床上常见的神经系统疾病,尤其在老年人中发病率日渐上升。Tau蛋白异常表达可导致认知功能障碍的发生,并常与其严重程度相关。文章概述Tau蛋白和认知功能障碍的发病关系、机制及临床应用,就Tau蛋白导致认知功能障碍的多种翻译后修饰、Tau蛋白传播机制、以Tau蛋白为治疗靶点的神经保护策略以及Tau蛋白正电子发射断层显像(positron emission tomography, PET)成像新技术进行综述。Tau蛋白导致认知功能障碍的机制研究可为认知功能障碍的诊断和治疗提供新思路。 Cognitive dysfunction is a common clinically neurological disease, especially with a high incidences in the elderly patients. Abnormal expression of Tau protein can cause the occurrence of cognitive dysfunction and is often associated with the severity of illness. This article overviews the relationship between Tau protein and cognitive dysfunction as well as mechanisms and clinical applications of Tau protein leading to cognitive dysfunction. We reviewed research advances on a variety of post-translational modifications of Tau, propagation mechanisms of Tau protein, neurological protection strategies targeting at Tau protein and Tau positron emission computed tomography (PET) imaging technology. The mechanisms of Tau protein leading to cognitive dysfunction can provide new implications on diagnosis and treatment of cognitive dysfunction.
