丹参酚酸B对过氧化氢引起的黑素细胞凋亡的保护作用研究 被引量:3

摘要 目的:研究丹参酚酸B对过氧化氢(H2O2)诱导的人表皮黑素细胞氧化应激及凋亡的保护作用。方法:采用MTT法检测H2O2对黑素细胞的活力抑制作用及丹参酚酸B的保护作用;Annexin-V/PI双染流式细胞术检测H2O2诱导黑素细胞凋亡的作用及丹参酚酸B的保护作用;DCFH-DA法流式细胞术检测H2O2促进黑素细胞活性氧簇(ROS)生成的作用及丹参酚酸B的抗氧化效果。结果:250μmol/LH2O2作用于黑素细胞24h可明显降低细胞活力、增加凋亡。0.6~10μmol/L丹参酚酸B预处理1h可明显保护黑素细胞,减少H2O2诱导的黑素细胞活力降低及凋亡。H2O2作用2h可显著诱导ROS生成,而丹参酚酸B可显著抑制H2O2诱导的ROS生成。结论:丹参酚酸B可能抑制活性氧生成,减少H2O2引起的黑素细胞凋亡,因而可能具有成为治疗白癜风新药的前景。 Objective To test the effects of magnesium tanshinoate B on H2O2-induced oxidative stress and apoptosis in human epidermal melanocytes. Methods Effects of H2O2 and magnesium tanshinoate B on cell viability were tested by MTT assay.The apoptosis of melanocytes were detected by annexin-V/PI staining cytometry analysis. ROS generation was measured by cytometry analysis with DCFH-DA method. Results The cell viability of melanocytes deceased and apoptosis rate and ROS generation increased after treated with 250μmol/L H2O2 for 24h, while pretreatment with 0.6~10μmol/L magnesium tanshinoate B significantly alleviated the effects of H2O2. Conclusions Magnesium tanshinoate B alleviated H2O2-induced apoptosis in melanocytes probably by inhibiting the ROS generation according to its antioxidant ability, thus may have the potential to be used in treating vitiligo in the future.
