3%七氟烷对老年大鼠认知功能的影响 被引量:1

摘要 目的 探讨吸入3%七氟烷对老年大鼠认知功能的影响。方法 30只老年SD雄性大鼠随机分为对照组(给予吸入30%氧气6 h)和S1组(给予3%七氟烷,30%氧气吸入6 h,1 d后行实验),S2组(给予3%七氟烷,30%氧气吸入6 h,7 d后行实验),每组各10只。分别在大鼠苏醒后1 d或7 d进行水迷宫实验,观察大鼠行为学变化。将大鼠脑组织切片,记录并比较单位长度海马神经元树突棘个数。通过Western Blot法测定大鼠海马NMDA受体NR2A亚基、NR2B亚基的表达。结果 S1组与对照组比较逃逸潜伏期延长、穿越平台次数减少(P<0.05)。S2组组逃逸潜伏期、穿越平台次数与对照组比较,差异均无统计学意义(P>0.05)。麻醉组单位长度下海马神经元的树突棘数量与对照组相比差异均无统计学意义(P>0.05)。S1组与对照组比较NR2A和NR2B表达明显下调(P<0.05)。结论 3%七氟烷造成麻醉后1 d老年大鼠认知功能障碍,但不造成麻醉后7 d老年大鼠认知功能障碍。 Objective To discuss the effects of 3% sevoflurane inhalation on cognitive function of aged rats. Methods Thirty healthy aged SD rats were randomly divided into group C(inhaled 30% oxygen for 6 hours as control), group S1 (inhaled 3%sevoflurane and 30%oxygen for 6 hours, be tested after inhaled 3%sevoflurane 1 day) and group S2 (inhaled 3% sevoflurane and 30% oxygen for 6 hours, be tested after inhaled 3% sevofluane 7 days) with 10 rats each. The changes of rats behavior were tested by Morris water maze within 1 or 7 days after the inhalation. Slice the rat brain, the number of hippocampal dendritic spines per unit length was recorded and compared. The expression of NR2A and NR2B were tested by WesternBlot. Results Morris Watermaze test demonstrated that the escape latency was longer and the number of crossing hidden-platform was significantly decreased in S1 group than those in control group (< 0.05). There was no significant difference between group C and group S2 in escape latency,number of crossing hidden-platform. There was no significant difference among group C,group S1 and group S2 in the number of hippocampal dendritic spines per unit length. The expression levels of NR2A and NR2B in group S1 were fewer than those in control group (<0.05). Conclusion 3% sevoflurane inhibits cognitive function of aged rats one day after anesthesia,but it does not cause cognitive dysfunction in aged rats one week after anesthesia.
